摘 要:【目的】开发水晶冰菜总黄酮的营养利用价值,并为纯天然植物抗氧化剂的开发提供理论基础。【方法】以水晶冰菜为原料,经反复冻融处理后采用超声辅助法提取总黄酮,再分别用石油醚、乙酸乙酯、正丁醇分级萃取以获得相应组分,测定不同组分中总黄酮的含量。以维生素C做阳性对照,测定各组分对DPPH·、·OH与O_(2)^(-)·的清除率。【结果】正丁醇和乙酸乙酯萃取组分中总黄酮含量分别为191.0 mg/g、184.5 mg/g,抗氧化活性显著高于其他组分;而且不同萃取组分在三种反应体系中也表现出不同程度的自由基清除能力,其中正丁醇萃取组分对DPPH·、·OH的清除能力最强,IC_(50)值分别是0.013 mg/mL、0.049 mg/mL,而乙酸乙酯萃取组分对O_(2)^(-)·的清除作用最强,IC_(50)值为0.156 mg/mL。【结论】水晶冰菜总黄酮在正丁醇和乙酸乙酯萃取组分中的含量高,具有良好的抗氧化活性,有望开发成为纯天然植物抗氧化剂。[Objective]To study the content of total flavonoids and antioxidant activity of total flavonoids in different components after fractional extraction with different polar solvents,and to clarify the relationship between them.[Methods]The total flavonoids were extracted by ultrasonic-assisted method after repeated freeze-thaw treatment.Petroleum ether,ethyl acetate and n-butanol were used to extract different components,and the mass concentration of total flavonoids in different components was determined.The scavenging rates of DPPH·,·OH and O_(2)^(-)·were determined by using vitamin C as positive control.[Results]The contents of flavonoids in n-butanol and ethyl acetate fractions of crude extract were 191.0 mg/g and 184.5 mg/g,respectively,and the antioxidant activity was significantly higher than that of other fractions.In addition,the antioxidant activities of different extraction components in each reaction system were also different.Among them,n-butanol had the strongest scavenging activity relative to DPPH and·OH,with IC_(50) value of 0.013 mg/mL and 0.049 mg/mL,respectively,while ethyl acetate had the strongest scavenging activity relative to O_(2)^(-)·,with IC_(50) value of 0.156 mg/mL.This study can not only improve the nutritional value of frozen vegetables,but also provide a theoretical basis for the development of natural plant antioxidants.[Conclusion]Mesembryanthemum crystallinum flavone has high content in n-butanol and ethyl acetate extraction phase and has good antioxidant activity,which is expected to be developed as a pure natural plant antioxidant.
