摘  要:新城疫病毒(NDV)属于单股负链病毒目(Mononegavirales)副粘病毒科(Paramyxoviridae)禽副粘病毒属(Avulavirus)。核不均一核糖体蛋白A1(hnRNPA1)在细胞核结合并影响前体mRNA的剪切和成熟mRNA的转运。为研究hnRNPA1是否调控NDV感染,本研究通过间接免疫荧光实验检测了hnRNAP1在病毒感染过程中的亚细胞定位。结果显示,NDV感染促进hnRNPA1蛋白出核,并与病毒核衣壳蛋白NP共定位;免疫共沉淀实验证实,hnRNPA1分别与病毒核衣壳蛋白NP和病毒膜蛋白M相互作用;以siRNA干扰hnRNPA1的表达,NDV增殖受到抑制,过表达hnRNPA1则促进NDV增殖。上述结果表明,NDV感染促进部分hnRNPA1进入细胞质,与病毒NP蛋白相互作用并促进病毒增殖。该研究结果为hnRNPA1参与副粘病毒复制的研究奠定了理论基础。Newcastle disease virus(NDV)belongs to the genus Avulavirus of the family Paramyxoviridae in the order Mononegavirales.Heterogeneous nuclear ribonucleoprotein A1(hnRNPA1)interacts with pre-mRNAs in the nucleus and is involved in the processing of pre-mRNA and transport of mRNA.To investigate whether hnRNPA1 regulated NDV infection,we checked the subcellular localization of hnRNPA1 upon NDV infection.We found that NDV infection induced hnRNPA1 nuclear export as shown in indirect immunofl uorescence assay.The cytoplasmic hnRNPA1 co-localized well with viral nucleocapsid protein NP,which was located to the virus replication complex.Co-immunoprecipitation experiments demonstrated that hnRNPA1 interacted with viral protein NP and M,respectively.When the expression of hnRNPA1 was interfered by using siRNA,NDV proliferation were suppressed whereas overexpression of hnRNPA1 promoted virus proliferation.These results suggested that during NDV infection,a proportion of hnRNPA1 was exported from nucleus to the cytoplasm and bound to viral protein NP,promoting viral RNA synthesis and increasing virus replication.This study shed light for the study of hnRNPA1 involved in paramyxovirus replication.
