摘 要:目的研究甘木通中10种活性化合物对H_(2)O_(2)诱导形成的H9C2心肌细胞损伤模型的保护作用。方法采用DPPH法测定甘木通活性化合物对自由基清除活性,铁还原法评价铁还原活性。用甘木通提取物处理H_(2)O_(2)诱导H9C2细胞建立的细胞损伤模型。MTT法测定细胞存活率,流式细胞术检测细胞凋亡率和ROS水平,Western blot法测定细胞凋亡相关蛋白半胱氨酸天冬氨酸蛋白酶3(caspase-3)、血红素氧合酶-1(HO-1)、半胱氨酸连接酶催化亚基(GCLC)表达水平。结果甘木通活性化合物能提高自由基清除活率,增加还原能力,其中木犀草素具有最高的活性。这些化合物对H_(2)O_(2)诱导的H9C2细胞氧化应激具有明显的抑制作用,降低细胞中活性氧的积累,提高抗氧化酶HO-1和GCLC的表达。结论甘木通活性化合物可不同程度改善由H_(2)O_(2)诱导的心肌细胞损伤,发挥心肌细胞保护作用,与其调节氧化应激的作用有关。Aim To investigate the protective effects of the 10 compounds from Clematis filamentosa Dunn.on H_(2)O_(2)-induced oxidative stress injury in rat myocardial cells.Methods DPPH,reducing power and MTT assays were then performed.The levels of reactive oxygen species and apoptosis rate were detected by flow cytometry.The levels of caspase-3 and antioxidant enzymes were analyzed by Western blot.Results DPPH and reducing power assays indicated the radical scavenging potential of these samples,and luteolin possessed the highest activity.These compounds had obvious inhibitory effects on oxidative stress induced by H_(2)O_(2)in H9C2 cells.They reduced the accumulation of ROS and improved the expression of antioxidant enzymes HO-1 and GCLC.Conclusion The 10 compounds extracted from Clematis filamentosa Dunn.can inhibit oxidative stress injury of H9C2 cells to different degrees and play protective roles in myocardial cells.
