锡粉促进下喹喔啉酮类化合物的二氟甲基化反应研究

摘  要:探索了在锡粉促进下,以溴二氟乙酸乙酯作为二氟亚甲基合成砌块,由喹喔啉-2(1H)-酮"一锅法"合成3-二氟亚甲基-3,4-二氢喹喔啉-2(1H)-酮类衍生物。反应的最优条件为:78℃回流;喹喔啉-2(1H)-酮,溴二氟乙酸乙酯,锡粉的摩尔比为1:2:2.5,添加剂为三氟化硼乙醚(2倍当量),溶剂为四氢呋喃。该法为合成3-二氟亚甲基-3,4-二氢喹喔啉-2(1H)-酮类化合物提供了一种新方法。“One-pot”synthesis of 3-difluoromethyl-3,4-dihydroquinoxaline-2(1H)-ones was explored using ethyl bromodifluoroacetate as difluoromethylation building block promoted by tin powder.The optimum reaction conditions were as following:reflux at 78℃,the molar ratio of quinoxaline-2(1H)-one,ethyl bromodifluoroacetate and tin powder was 1􀏑2􀏑2.5,the additive was BF3·Et_(2)O(2 equivalent),and the solvent was tetrahydrofuran.The method provided a new method for the synthesis of 3-difluoromethyl-3,4-dihydroquinoxaline-2(1H)-one derivatives.
