摘 要:目的在心血管基础研究中,麻醉剂能减少实验动物的痛苦,但会影响心率。本文探讨如何避免麻醉对动物心电图记录的影响。方法16只清洁级SD大鼠,随机分为麻醉组及清醒组,每组8只。麻醉组大鼠给予4%水合氯醛10 mg/kg进行腹腔注射,清醒组以等量生理盐水腹腔注射。麻醉组动物达到理想麻醉状态30 min后将清醒组和麻醉组大鼠均按照30 mg/kg体重的剂量进行腹腔异丙肾上腺素注射。应用掌上心电E-U08记录大鼠清醒状态下的心电图,与多导生理仪记录的大鼠麻醉状态下心电图比较;比较两组大鼠注射异丙肾上腺素后的心率、ST段变化。结果掌上心电记录到的大鼠清醒状态下的心电图与多导生理仪记录到的大鼠麻醉状态下的心电图心率、波形一致性较好;大鼠注射异丙肾上腺素后诱发心肌缺血,掌上心电E-U08可较为准确记录到心肌缺血心电图,表现为ST段抬高,统一标尺下与多导生理仪记录的心电图一致。注药后1 min,清醒组大鼠心率(324.0±11.1)次/min高于麻醉组的(308.0±10.5)次/min,差异具有统计学意义(t=2.962,P<0.05)。结论掌上心电E-U08可应用于大鼠清醒时心率的实时记录。Objective In basic cardiovascular research,anesthetics can reduce pain in experimental animals but can affect heart rate.This article discusses how to avoid the effects of anesthesia on ECG recordings in animals.Methods A total of 16 clean-grade SD rats were randomly divided into anesthesia group and awake group,with 8 rats in each group.Rats in the anesthesia group were injected intraperitoneally with 4%chloral hydrate 10 mg/kg,and rats in the awake group were injected intraperitoneally with the same amount of normal saline.30 min after the animals in the anesthesia group reached the ideal anesthesia state,the rats in the awake group and the anesthesia group were injected intraperitoneally with isoproterenol at a dose of 30 mg/kg body weight.E-U08 Snap ECG recorder was used to record the ECG of the rat in the awake state,and compared it with the ECG of the rats under the anesthesia recorded by the polygraph system.The changes of heart rate and ST segment in two groups after injection of isoproterenol were compared.Results The ECG recorded by E-U08 Snap ECG recorder in the awake state of the rat is consistent with the heart rate and waveform of the ECG recorded by thepolygraph system in the anesthetized state of the rat.Myocardial ischemia was induced by the injection of isoproterenol in rats.E-U08 Snap ECG recorder can record myocardial ischemia ECG more accurately,showing ST-segment elevation,which is consistent with the ECG recorded by the polygraph system under the unified ruler.1 min after the injection,the heart rate of rats in the awake group was(324.0±11.1)beats/min,which was higher than(308.0±10.5)beats/min in the anesthesia group,and the difference was statistically significant(t=2.962,P<0.05).Conclusion E-U08 Snap ECG recorder could be used to record the heart rate of awake rats in real-time.
