摘 要:目的观察胰岛素对3个月和6个月糖尿病病程大鼠颌下腺超微结构的影响。方法SD雄性大鼠48只,16只作为对照组,32只给予高脂饲料喂养,腹腔注射链脲佐菌素,复制Ⅱ型糖尿病模型,选模成功后随机分为两组,其中16只给予胰岛素处理作为治疗组,另外16只作为糖尿病组。在造模后3个月和6个月每组各处死8只大鼠,腹腔采血检测空腹血糖(Fasting blood glucose,FBG)水平,并取颌下腺组织进行电镜观察。结果(1)空腹血糖:糖尿病组空腹血糖较同期对照组升高,且造模后6个月高于造模后3个月,差异均有统计学意义(P<0.05);治疗组空腹血糖较同期糖尿病组下降,较同期对照组升高,且造模后6个月高于造模后3个月,差异均有统计学意义(P<0.05)。(2)电镜观察:糖尿病组颌下腺颗粒曲管上皮出现线粒体断裂、空泡变性,腺细胞内粗面内质网呈扩张改变,并随着病程延长加重;治疗组在造模后6个月时病变轻微。结论糖尿病大鼠出现颌下腺超微结构改变并随病程的延长加重,而胰岛素能明显控制其结构改变。Objective To observe the effect of insulin on the ultrastructure of submandibular gland in diabetic rats with 3 months and 6 months course of disease.Methods 48 SD male rats, 16 rats served as control group, 32 rats were given high-fat diet, after producing insulin resistance, to copying type II diabetes model, 16 rats were treated with insulin as treatment group, and another 16 rats as diabetes group.After molding for 3 months and 6 months, half of the rats in each group were sacrificed.Sampling abdominal blood for fasting blood glucose detected, and the submandibular gland tissues were taken for electron microscopy observation.Results(1) Fasting blood sugar: Fasting blood glucose in the diabetes group was increased by comparing with the control group in the same period, after molding of 6 months was higher than after molding of 3 months, and the differences were statistically significant(P<0.05);Blood glucose in the treatment group was reduced by comparing with the diabetes groups in the same period, and was increased by comparing with the control group in the same period, after molding of 6 months was higher than after molding of 3 months, and the differences were statistically significant(P<0.05).(2)Electro microscopy observation: The mitochondrial of granule curved tubule epithelial layer of submandibular gland was broken and vacuolated, The rough endoplasmic reticulum in the gland cells expanded, and aggravated with the extension of process.As molding of 6 months, the lesion was mild in the treatment group.Conclusion Rat submandibular gland in the diabetes group has disorder ultrastructure, and aggravated with the extension of process.Insulin can significantly control the structural disorder of submandibular gland in diabetic rats.
