摘 要:目的探讨截短型转化生长因子βII型受体(tTβRII)对四氯化碳(CCl_(4))诱导C57BL/6J小鼠急性肝损伤炎症因子表达的影响。方法将20只C57BL/6J小鼠随机分为空白对照组、模型组、模型对照组和tTβRII处理组,每组5只。空白对照组小鼠腹腔注射200μL纯橄榄油,每天1次,共4 d;其余三组小鼠均腹腔注射200μL 40%的CCl_(4)/橄榄油混合液,1次/d,共4 d,同时模型对照组和tTβRII处理组小鼠分别在第3天和第4天尾静脉注射PBS和目的蛋白tTβRII(60μg)。最后一次注射24 h后麻醉处死小鼠并取出肝组织,苏木素-伊红(HE)染色观察肝组织病理学变化;实时荧光定量PCR检测肝组织中炎症细胞因子TNF-α和IL-1βmRNA的表达;免疫组化染色检测肝组织中TNF-α和IL-1β蛋白的表达。结果(1)HE染色结果表明,与空白对照组相比,模型组中肝小叶排列紊乱,肝细胞发生变性坏死,经tTβRII处理后,肝小叶排列趋于规则且肝细胞形态趋于完整。(2)实时荧光定量PCR表明,与空白对照组相比,模型组中TNF-α和IL-1βmRNA表达均显著增加(F=202.322,P=5.5664E-8;F=72.595,P=0.000002),而与模型组相比,tTβRII处理组中TNF-α和IL-1βmRNA表达均显著下降(F=202.322,P=0.000001;F=72.595,P=0.007624)。(3)免疫组化染色表明,与空白对照组相比,模型组中TNF-α和IL-1β蛋白表达均上调,而与模型组相比,tTβRII处理组中TNF-α和IL-1β蛋白表达均下调。结论目的蛋白tTβRII对CCl_(4)诱导的急性肝损伤具有一定的保护作用,其作用可能是通过影响炎症因子表达实现的。Objective To investigate the effect of truncated transforming growth factor β receptor type II on the inflammation responses in carbon tetrachloride-induced acute liver injury in mice.Methods C57 BL/6 J mice were divided into blank control, model, model control and tTβRII treatment groups.Mice in the blank control group received anintraperitoneal injection of pure olive oil(200 μL) once a day for 4 days, while mice in model, model control and tTβRII treatment groups received intraperitoneal injection of carbon tetrachloride(200 μL) once a day for 4 days.Meanwhile, mice in model control and tTβRII treatment groups were treated intravenously with PBS and tTβRII(60 μg/mouse) at day 3 and 4.All mice were sacrificed, and then liver tissues were collected.The histopathology of liver tissues in different groups was evaluated by hematoxylin and eosin(HE) staining.The mRNA expression levels of TNF-α and IL-1β were detected by real-time PCR.The protein expression levels of TNF-α and IL-1β were detected by immunohistochemistry staining.Results(1) The disorder of hepatic lobules and the necrosis of hepatocytes in the model group were observed relative to that in the blank control group, while we observed the regular arrangement of hepatic lobules and the intact morphology of hepatocytes in tTβRII treatment group.(2) The mRNA expression levels of TNF-α and IL-1β were increased in the model group compared with the control group(F=202.322,P=5.5664E-8;F=72.595,P=0.000002),and the mRNAs of TNF-α and IL-1β were inhibited in the tTβRII-treated model group(F=202.322,P=0.000001;F=72.595,P=0.007624).(3) The protein expression levels of TNF-α and IL-1β were upregulated in the model group compared with the control group.The protein expression levels of TNF-α and IL-1β were downregulated in the tTβRII-treated model group.Conclusion The protein tTβRII protects against CCl4-induced acute liverinjury in mice, which may be associated with the inflammatory responses.
