摘  要:以早熟苹果优良新品种‘鲁丽’无菌苗的离体叶片的外植体为试材,采用离体组织培养方法,研究了植物生长调节物质及碳源对叶片高效不定芽再生的影响,以期获得诱导‘鲁丽’叶片高效不定芽再生的适宜培养基组成。结果表明:不依赖于植物生长调节物质的组成,碳源D-山梨醇相比蔗糖明显提高不定芽再生率和平均单位叶片不定芽数。在相同碳源条件下,不定芽再生率和单位叶片不定芽数均表现为塞苯隆(TDZ)高于6-苄基氨基嘌呤(BA),但TDZ和BA之间没有达到差异显著水平;TDZ及BA不同浓度间差异不显著。诱导不定梢绿苗生根,除了较高浓度1.0 mg·L^(-1)IBA处理外,基本培养基1/4MS比1/2MS显著提高不定梢生根率;在基本培养基1/4MS上,生长素物质吲哚乙酸(IAA)比吲哚丁酸(IBA)显著提高生根率和单株生根数。在基本培养基1/2MS上,IAA和IBA的生根率和单株生根数均差异不显著。IAA比IBA诱导生根速度快,IAA诱导的不定根从绿苗茎横切面直接产生,没有愈伤产生,最高生根率为80.8%,平均单株生根数为4.2条。New expanding leaves from in vitro shoot top of new varieties‘Luli’(Malus domestica var.‘Luli’)with early mature and good quality were selected as materials.The effects of plant growth regulators and carbon source on adventitious bud regeneration from leaf explants were examined.The results showed that carbon source D-sorbitol than sucrose markedly improved adventitious bud regeneration frequency and averaged bud number per leaf explant,independent of plant growth regulators.Under the same carbon source,both bud regeneration frequency and bud number per leaf explant showed TDZ was higher than BA,but didn’t reach significant difference between TDZ and BA.For in vitro rooting of adventitious shoots,basal medium 1/4 MS than 1/2 MS significantly improved rooting rate,exclusive of adding higher IBA concentration 1.0 mg·L^(-1).On basal medium 1/4 MS,auxin IAA than IBA significantly improved rooting rate and root number per plantlet.On basal medium 1/2 MS,both rooting rate and root number per plantlet showed no significant difference between IAA and IBA.But rooting process induced by IAA was faster than that by IBA.Roots induced by IAA were directly produced from stem cross without callus production.The highest rooting rate was 80.8%,and root number per plantlet was 4.2.
