摘 要:目的:研究淫羊藿苷对少精子症模型大鼠生殖功能的保护作用及可能的作用机制。方法:将60只雄性SD大鼠按体质量随机分为正常组、模型组、男宝胶囊组和淫羊藿苷低、中、高剂量组。各药物组大鼠灌胃给予相应药物,正常组及模型组大鼠灌胃给予等体积去离子水,1次/d,连续给药4周。除对照组外,其他组大鼠给药4 h后,灌胃给予腺嘌呤混悬液造模,正常组给予溶媒,1次/d,连续4周。以精子密度、精子畸形率、性器官指数、肌酐、尿素氮、血清睾酮含量和睾丸组织中SOD和MDA为检测指标观察各组大鼠的情况。结果:与模型组比较,淫羊藿苷能够明显增加少精子症模型大鼠精子密度、精子活动率和性器官指数,明显降低肾功能指标肌酐和尿素氮,升高血清睾酮含量(P<0.05或P<0.01);与模型组比较,淫羊藿苷组大鼠睾丸组织中SOD活性明显升高,MDA含量明显降低(P<0.01)。结论:淫羊藿苷对少精子症模型大鼠的生殖功能具有良好的保护作用,其机制可能与其抗氧化作用有关。Objective:To study the protective effect and the potential mechanism of icariin on the reproductive function of oligospermia model rats.Methods:A total of 60 male SD rats were randomly divided into normal group,model group,Nanbao capsule group,icariin low-dose group,icariin medium-dose group and icariin high-dose group according to their body weight.The rats in each drug group were given the corresponding drugs by gavage,and the rats in the normal group and the model group were given the same volume of deionized water by gavage,once a day,for 4 weeks.After 4 hours of administration every day,except the control group,rats in the other groups were given adenine suspension by intragastric administration,and the normal group was given the vehicle,once a day for 4 consecutive weeks.Sperm density,sperm motility,sex organ index,creatinine,urea nitrogen,serum testosterone content,and SOD and MDA in testicular tissue were used as test indicators to observe the conditions of rats in each group.Results:Compared with the model group,icariin can significantly increase sperm density,sperm motility and sexual organ index in oligospermia model rats,significantly reduce renal function indicators creatinine and urea nitrogen,and increase the content of serum testosterone(P<0.05 or P<0.01).Compared with the model group,the SOD activity in the testicular tissue of the icariin group was significantly increased,and the MDA content was significantly decreased(P<0.01).Conclusion:Icariin has a good protective effect on the reproductive function of oligospermia model rats,and its mechanism may be related to its antioxidant effect.
