摘 要:目的:基于"风痰入里成瘤说"理论探究中药猫眼草的抗癌作用及物质基础,研究中药猫眼草提取物的化学成分,为进一步研究其抗癌的活性成分提供依据。方法:采用正相硅胶柱、C18反相柱、高效液相色谱、SephadexLH-20等多种色谱手段对猫眼草95%乙醇提取物进行化学成分分离,并根据理化性质和波谱数据鉴定各化合物的结构。对分离所得化合物,采用HONE-1(未分化型人鼻咽癌细胞)、MCF-7(人乳腺癌细胞)和MGC-803(人胃癌细胞)进行抗癌活性评价。结果:从猫眼草中分离鉴定得到6个化合物,分别为:esulone A(1)、mesulones C(2)、kanesulone B(3)、supinenolone E(4)、supinenolone E acetate(5)和cycloart-(23Z)-ene-3β,25(6)。化合物4、5和6为首次从大戟属植物中分离得到。结论:三萜类化合物4、5和6具有潜在的抗鼻咽癌和乳腺癌活性(10μmol/L时的抑制率大于50%),其中化合物6活性最好,对HONE-1和MCF-7细胞抑制增殖活性分别为5.2μmol/L和6.6μmol/L。Objective:Based on the theory of"wind phlegm leading to tumor",this paper explores the effect and material basis of Maoyancao(Euphorbia Esula),in the treatment of cancer,and studies the chemical constituents extract of Maoyancao(Euphorbia Esula),so as to provide basis for further study of its anti-cancer active components.Methods:The chemical constituents of 95%ethanol extract of Maoyancao(Euphorbia Esula)were separated by Normal phase silica gel column,C18 reversed phase column,HPLC and sephadexlh-20,and their structures were identified according to their physicochemical properties and spectral data.The antitumor activities of the compounds were evaluated by cell models of HONE-1,MCF-7 and MGC-803.Results:Totally six compounds were isolated and identified as esulone A(1),Mesones C(2),kanesulone B(3),supinenolone E(4),supinenolone E acetate(5)and cycloart-(23 Z)-ene-3β,25.Compounds 4,5 and 6 are isolated from Euphorbia for the first time.Conclusion:Triterpenoids 4,5 and 6 have potential antitumor activities against nasopharyngeal carcinoma and breast cancer(the inhibition rate is more than 50%at 10μmol/L).Compound 6 has the best activity,and its inhibitory activity on HONE-1 and MCF-7 cells is 5.2μmol/L and 6.6μmol/L,respectively.
