摘  要:目的:基于网络药理学探索桑贝止嗽散治疗新型冠状病毒肺炎(Corona Virus Disease 2019,COVID-19)(简称新冠肺炎)的药理机制。方法:通过中药网络药理学平台(traditional Chinese medicine systems pharmacology database and analysis platform,TCMSP)检索桑贝止嗽散中桑白皮、浙贝母、百部、紫菀四味核心药物的主要活性成分及其对应的靶标基因,同时通过Gene Cards数据库获得新冠肺炎相关靶点,将两者进行映射,获得交集靶点作为桑贝止嗽散治疗新冠肺炎的预测靶点,利用Cytoscape软件、STRING和DAVID 6.8等数据库构建蛋白互作(protein protein interaction,PPI)网络,并进行基因本体(geneontology,GO)富集分析及基因组百科全书(kyoto encyclopedia of genes and genomes,KEGG)通路分析。结果:桑贝止嗽散4味核心药物作用于新冠肺炎的活性成分有68个,相关靶点87个,关键活性化合物8个,关键靶基因17个;GO分析显示,桑贝止嗽散治疗新冠肺炎影响的生物过程主要有脂多糖介导的信号通路,蛋白信号转导,缺氧反应等15个;KEGG通路富集显示桑贝止嗽散治疗新冠肺炎的通路主要有:PI3K-Akt信号通路,钙离子信号通路,T细胞受体信号转导,TNF信号通路等5条。结论:桑贝止嗽散可能通过以上相关通路作用于TNF、VEGFA等关键靶点,影响蛋白表达、信号通路传导等生物过程,发挥治疗新冠肺炎的作用,为实验研究提供一定的理论参考依据。Objective:To explore the pharmacological mechanism of Sangbei cough-stopping powder in treating COVID-19 based on network pharmacology.Methods:The main active ingredients of Sangbaipi(Cortex Mori),Zhebeimu(Bulbus Fritillariae thunbergii),Baibu(Japanese Stemona Root)and Ziwan(Aster tongolensis)contained in Sangbei cough-stopping powder and their corresponding target genes were searched via TCMSP,simultaneously,the related targets of COVID-19 were obtained by Gene Cards database,both were mapped to obtain the intersection target as the prediction targets of Sangbei cough-stopping powder in treating COVID-19,Cytoscape software,STRING and DAVID 6.8 were utilized to construct PPI network,and carry out GO analysis and KEGG pathway analysis.Results:Among four core drugs contained in Sangbei cough-stopping powder,there were 68 active ingredients acting on COVID-19,87 related targets,eight key active compounds and 17 key target genes;GO analysis showed that the treatment of COVID-19 with Sangbei cough-stopping powder mainly involved 15 biological processes,including lipopolysaccharide mediated signal pathway,protein signal transduction and hypoxia response;KEGG pathway analysis displayed that the pathway of the treatment of COVID-19 with Sangbei cough-stopping powder contained PI3K-Akt signaling pathway,calcium signaling pathway,T cell receptor signal transduction,TNF signaling pathway and others.Conclusion:Sangbei coughstopping powder develop the mechanism of treating COVID-19 possibly by acting on key targets such as TNF,VEGFA and other key targets through the above related pathways and affecting biological processes such as protein expression and signal pathway transduction,and it could provide a certain theoretical reference for experimental research.
