人参皂苷Rb_(1)对兔离体肠平滑肌收缩活动的影响

摘  要:目的:研究人参皂苷Rb_(1)对兔离体肠平滑肌收缩活动的影响。方法:采用兔离体肠平滑肌实验,应用BL-420F生物机能实验系统,观察人参皂苷Rb_(1)对正常状态下兔离体肠平滑肌自发性收缩的影响,探究人参皂苷Rb_(1)影响兔离体肠平滑肌收缩的作用机制。结果:人参皂苷Rb_(1)能浓度依赖性地抑制兔离体肠自发性收缩,且浓度在70、90µmol/L可明显降低兔离体小肠平滑肌自发性收缩张力和收缩振幅(P<0.01),但对频率无影响(P>0.05)。用钌红、肝素和维拉帕米对兔离体肠平滑肌预处理后,人参皂苷Rb_(1)对兔离体肠平滑肌收缩的抑制程度明显增强(P<0.05)。结论:人参皂苷Rb_(1)可显著抑制兔离体肠平滑肌自发性收缩,其机制可能是阻断L-型Ca^(2+)通道抑制外钙内流,或阻断ryanodine受体和三磷酸肌醇(inositol triphosphate,IP3)受体抑制内钙释放,最终使细胞内钙离子浓度降低,从而抑制肠平滑肌的收缩。Objective:To study the effects of ginsenoside Rb_(1) on the contraction of isolated rabbit intestine smooth muscle.Methods:The experimental study on the isolated rabbit intestinal smooth muscle,and BL-420F biological function experiment system were applied to observe the effects of ginsenoside Rb_(1) on spontaneous contraction of the isolated rabbit intestinal smooth muscle under normal state,and explore the mechanism of ginsenoside Rb_(1) acting on the contraction of isolated rabbit intestine smooth muscle.Results:Ginsenoside Rb_(1) could inhibit spontaneous contraction of the isolated rabbit intestinal smooth muscle in dose-dependent manner,it could reduce spontaneous contractile tension and amplitude of isolated small intestinal smooth muscle in rabbits in the concentrations of 70 and 90μmol/L notably(P<0.01),while it showed no effects on the frequency(P>0.05).After preconditioning the isolated rabbit intestinal smooth muscle with ruthenium red,heparin and verapamil,the inhibitory degrees of ginsenoside Rb_(1) enhanced notably on the contraction of isolated rabbit intestine smooth muscle(P<0.05).Conclusion:Ginsenoside Rb_(1) could significantly inhibit spontaneous contraction of the isolated rabbit intestinal smooth muscle,and its mechanism may be inhibiting calcium influx by blocking L-type Ca^(2+) channel,or inhibiting the release of intracellular calcium by blocking ryanodine receptor and IP3 receptor,eventually reduces the concentration of intracellular calcium,thereby inhibiting the contraction of intestinal smooth muscle.
