生肌玉红膏对大鼠糖尿病足溃疡组织bFGF、HIF-1α及VEGF表达的影响

摘　　要：目的:探讨生肌玉红膏对大鼠糖尿病足溃疡组织碱性成纤维细胞生长因子(basic fibro-blast growth factor,bFGF)、低氧诱导因子1α(hypoxia inducible factor-1α,HIF-1α)及内皮生长因子(vascular endothelial growth factor,VEGF)表达的影响。方法:选用SD大鼠50只制备糖尿病足模型,造模成功后将大鼠随机分为对照组和实验组各25只。大鼠足部溃疡处对照组覆盖凡士林纱布,实验组覆盖生肌玉红膏油纱布,隔日更换1次;分别于造模后3、7、14天取大鼠足创面局部组织标本,运用免疫组化法分别观察bFGF、HIF-1α及VEGF表达水平。结果:干预后实验组大鼠足创面局部组织中bFGF、HIF-1α及VEGF表达均高于对照组(P<0.05),其中第3天,HIF-1α表达两组比较差异有统计学意义(P<0.01),第7天,VEGF表达实验组高于对照组(P<0.01)。实验组bFGF、HIF-1α表达呈逐渐下降趋势,VEGF表达呈先升高后下降趋势。结论:生肌玉红膏能够促进创面局部组织中bFGF、HIF-1α及VEGF的表达,促进血管生成,改善糖尿病足溃疡缺血缺氧状态,缩短创面愈合时间,较凡士林有明显优势。Objective:To explore the effects of Shengji Yuhong ointment on the expressions of bFGF,HIF-1αand VEGF in ulcer tissue of the rats suffering diabetic foot.Methods:All 50 SD rats were chosen to prepare animal models with diabetic foot,after successful modeling,the rats were randomized into the control group and the experiment group,25 rats in each group.The control group were given vaseline at the foot ulcer,and the experiment group Shengji Yuhong ointment,changing once every other day;the samples of rat foot were taken at the 3rd,7th and 14 th day after modeling,immunohistochemical method was used to observe the expressions of bFGF,HIF-1αand VEGF respectively.Results:After the intervention,the expressions of bFGF,HIF-1αand VEGF of the experiment group were higher than these of the control group(P<0.05),the difference had statistical meaning between both groups in the expressions of HIF-1αat the 3rd day(P<0.01),the expressions of VEGF of the experiment group were higher than these of the control group at the 7th day(P<0.01).The expressions of bFGF and HIF-1αin the experiment group showed a gradual downward trend,while the expressions of VEGF increased first and then decreased.Conclusion:Shengji Yuhong ointment could lift the expressions of bFGF,HIF-1αand VEGF in local tissue of the wound,promote angiogenesis,improve the ischemia and hypoxia in diabetic foot ulcer,shorten wound healing time,it shows the notable advantage compared with vaseline.
