海南岛索氏桃花水母的遗传学鉴定

摘　　要：索氏桃花水母是一类广布于除南极洲外全球各地的腔肠动物,生长于天然或人工的各种淡水生境中,在我国已有多例报道,主要以长江流域较多。对2019年在海南儋州大王岭水库发现的淡水水母提取COX1和ITS基因并测序,经序列相似性比对后认为样品为索氏桃花水母(Craspedacusta sowerbyi Lankester,1880),并与其他发表的索氏桃花水母的相应基因序列共同构建系统进化树,发现在COX1基因上,海南儋州的桃花水母与美国的索氏桃花水母(LN901194.1)聚为一支,在ITS基因上与中国湖北的索氏桃花水母(AY513628、AY513626)聚为一支。在这之前,未见海南岛正式发现索氏桃花水母的学术报道,就所查文献也未见我国在海岛上发现该物种的正式报道。Craspedacusta sowerbyi Lankester,1880,freshwater jellyfish distributing all over the world except Antarctica,adapts to both natural and artificial freshwaters.There are many reports on the discovery of Craspedacusta in China,especially in Yangtze River Basin.On the freshwater jellyfish discovered in Dawangling Reservoir,Danzhou,Hainan in 2019,a molecular identification was conducted.After COX1 and ITS genes of the collected samples were extracted and sequenced,the species were identified as Craspedacusta sowerbyi through sequence alignment.After the phylogenetic tree was mapped with other published Craspedacusta sowerbyi COX1 and ITS sequence,the results showed that Danzhou sample clusteres with an American sample(LN901194.1)on COX1 gene and samples from Hubei,China(AY513628,AY513626).Before the current discovery,there was no academic report on the official discovery of Craspedacusta sowerbyi in Hainan Island,and no official report on the discovery of this species on any island in China.
