摘  要:目的:探讨艾灸“关元”穴对衰老进程中大鼠子宫组织细胞周期相关蛋白CyclinD1、CDK4、E2F1、SOD和CAT含量的影响,进一步探究艾灸延缓子宫衰老的机制。方法:选取SPF级12月龄Wistar雌性大鼠14只,随机分为对照组与艾灸组,SPF级3月龄Wistar雌性大鼠7只作为青年组。其中艾灸组大鼠对“关元”穴进行20 min温和灸刺激,每日1次,连续治疗40 d;对照组和青年组大鼠进行同样的抓取与固定。实验结束后取大鼠子宫组织,以酶联免疫吸附测定(ELISA)法检测细胞周期相关蛋白CyclinD1、CDK4、E2F1、SOD和CAT含量。结果:与青年组比较,对照组大鼠子宫组织中SOD、CAT、CyclinD1、CDK4和E2F1含量降低,差异具有统计学意义(P<0.05);而与对照组比较,艾灸组上述蛋白含量明显上升,差异具有统计学意义(P<0.05);各组大鼠子宫组织中CyclinD1含量与SOD、CAT含量均无相关性(P>0.05)。结论:艾灸“关元”穴可上调衰老进程中大鼠子宫组织中SOD、CAT含量,提高其抗氧化能力;可调节子宫组织细胞周期相关蛋白CyclinD1、CDK4的表达,促进E2F1的释放,使细胞加速通过G1期,进而对抗细胞衰老。艾灸的抗衰老作用可能是通过以上两条途径分别进行的。Objective:To investigate the effect of moxibustion at Guanyuan(CV4)on the contents of CyclinD1,CDK4,E2F1,SOD and CAT in uterine tissues of aging rats,thus to further explore the mechanism of moxibustion in delaying uterine senescence.Methods:14 SPF grade female Wistar rats of 12 month old were randomly divided into the control group and the moxibustion group.7 SPF grade female Wistar rats of 3 months old were selected as the young group.The rats in the moxibustion group were treated with mild moxibustion at ST25 for 20 minutes,once a day for 40 days;however,the control group and the young group were intervened with the same grasping and fixation.At the end of the experiment,the contents of cell cycle-related proteins(CyclinD1,CDK4,E2F1)and antioxidant enzymes(SOD,CAT)in uterine tissues were detected by enzyme-linked immunosorbent assay(ELISA).Results:Compared to those in the young group,the levels of SOD,CAT,CyclinD1,CDK4 and E2F1 were significantly decreased in the control group(P<0.05);which were significantly increased in the moxibustion intervention group when compared to those in the natural aging group(P<0.05).There was no correlation between the level of CyclinD1 and the levels of SOD and CAT in the uterine tissues of the rats in each group(P>0.05).Conclusion:Moxibustion at CV4 can up-regulate the levels of SOD and CAT in uterine tissues of female natural aging rats,strengthen their antioxidant capacity,regulate the expressions of cell cycle-related proteins of CyclinD1 and CDK4,promote the release of E2F1,accelerate the cell through G1 phase,thus to resist cellular aging.The anti-aging effect of moxibustion may be through the above two ways.
