摘 要:目的:探讨头穴丛刺调控NF-κB信号通路介导阿尔茨海默病(AD)模型大鼠炎性反应的作用机制。方法:将SPF级雄性Wistar大鼠随机分为正常组、假手术组、模型组和头穴丛刺组,每组12只。采用双侧海马内注射Aβ1-42制备AD模型。造模成功后第7天开始干预,头穴丛刺组选取百会穴及百会穴左侧旁开1 mm处、百会穴右侧旁开1 mm进行针刺干预,1次/d,留针30 min,连续干预14 d。采用免疫组化法检测海马组织COX-2、NF-κB的表达;采用实时荧光定量PCR法检测海马组织IL-1β、IL-6及TNF-αmRNA的表达。结果:与假手术组比较,模型组大鼠海马组织COX-2、NF-κB表达量显著升高(P<0.01);与模型组比较,头穴丛刺组大鼠海马组织COX-2、NF-κB表达量明显降低(P<0.01)。与假手术组比较,模型组大鼠海马组织IL-1β、IL-6及TNF-α基因mRNA表达水平均显著升高(P<0.01);与模型组比较,头穴丛刺组大鼠海马组织IL-1β、IL-6及TNF-α基因mRNA表达水平均明显降低(P<0.01)。结论:头穴丛刺针刺法调控NF-κB信号通路,抑制炎性因子表达,发挥其抗炎作用。Objective:To investigate the role and mechanism of scalp clustering acupuncture in regulating NF-κB signaling pathway intervening inflammatory response in the model rats with Alzheimer’s disease(AD).Methods:SPF grade male Wistar rats were randomly divided into the normal group,the sham operation group,the model group and the scalp clustering acupuncture group,with 12 rats in each group.AD model was established by bilateral injection of Aβ1-42 into hippocampus.The intervention began on 7 d after successful modeling,the points chose in the scalp clustering acupuncture group were Baihui(DU20),points 1mm left and right lateral to DU20,once per day for 14 continuous days.The expressions of COX-2 and NF-κB of hippocampal tissues were detected by immunohistochemistry,and the mRNA expressions of IL-1β,IL-6 and TNF-αwere measured by real-time fluorescence quantitative PCR.Results:The expressions of COX-2 and NF-κB were significantly increased in hippocampal tissues in the model group compared to those in the sham operation group(P<0.01);which were significantly decreased after the intervention of scalp clustering acupuncture(P<0.01).The mRNA expressions of IL-1β,IL-6 and TNF-αwere significantly increased in hippocampal tissue in the model group compared to those in the sham operation group(P<0.01);which were significantly decreased after the intervention of scalp clutering acupuncture(P<0.01).Conclusion:Scalp clustering acupuncture can regulate NF-κB signaling pathway and inhibit the expressions of inflammatory factors to exert its anti-inflammatory effect.
