摘  要:bZIP转录因子在植物的生长发育和响应胁迫的过程中起重要作用。本研究利用全长均一化cDNA文库获得一条全长438 bp cDNA序列,与香蕉基因组数据库比对之后,将其命名为MabZIP53,编码145个氨基酸,预测分子量为35.24 kD,等电点为5.22,其编码的蛋白定位于细胞核。系统发育分析表明MabZIP53与凤梨(XP020114228.1)、大叶藻(KMZ62676.1)的亲缘关系较近。MabZIP53在不同非生物胁迫(低温,干旱,盐)下均上调表达,尤其干旱胁迫下相对表达量最为显著。在接种Foc TR4后,抗病品种‘GCTCV-119’中MabZIP53表达量显著上调,而感病品种巴西蕉中MabZIP53表达量下调表达。这些研究结果表明MabZIP53可能参与香蕉响应生物胁迫和非生物胁迫的过程,为香蕉的遗传改良提供理论基础。bZIP transcription factors(basic leucine zipper, bZIP) play important roles in plant growth, development and stress response. In this study, we obtain a 438 bp cDNA sequence using full-length homogeneous cDNA library sequence, which encode a full-length bZIP transcription factor gene. After blasted in NCBI and in banana genome database, the sequence was named MabZIP53, encoding 145 amino acids, the predicted molecular weight and isoelectric point were 35.24 kD and 5.22, respectively. MabZIP53 proteins are localized in the nucleus(Nucleus). Phylogenetic analysis showed that the relationship between MabZIP53 and Ananas comosus(XP020114-228.1) and Zostera marina(KMZ62676.1) was closely. Moreover, MabZIP53 were up-regulated under different abiotic stresses(low temperature, drought, salt), and the relative expression of MabZIP53 was the most significant under drought stress. The expression of MabZIP53 was significantly up-regulated in the GCTCV-119 of resistant varieties inoculated with Foc TR4, and down-regulated in the Cavendish of susceptible varieties, indicating that MabZIP53 might participate in the process of responding to biotic and abiotic stresses in bananas. This study provide a theoretical basis information for genetic improvement of banana.
