努比亚山羊POU1F1与FOLR1基因多态性及其与断奶重的关联分析

摘  要:为了探索努比亚山羊垂体特异性转录因子1(POU1F1)与叶酸受体α基因(FOLR1)的基因多态性及其与断奶重的关联性,试验以355只努比亚山羊为研究对象,利用DNA混池法和Sanger测序法检测2个基因的候选位点;应用MTA-seq测序技术对单核苷酸多态性(SNPs)位点进行基因分型;根据分型统计结果检测Hardy-Weinberg平衡并计算群体遗传参数;利用GLM模型分析基因多态性及其与努比亚山羊断奶重的关联效应。结果表明:在POU1F1基因中检测到1个多态性位点POU1F1:g.34236170A>G,包含AA、AG、GG 3种基因型,其中AA为优势基因型,A为优势等位基因。在FOLR1基因中共检测到2个SNPs位点,其中FOLR1:g.31625869T>G位点包含TT、TG、GG 3种基因型,GG为优势基因型,G为优势等位基因;FOLR1:g.31626833G>A位点包含GG、GA、AA 3种基因型,其中GG为优势基因型,G为优势等位基因。POU1F1:g.34236170A>G位点和FOLR1:g.31625869T>G位点处于中度多态(0.25A位点杂合度较低,属于低度多态(PIC<0.25),这3个位点均达到Hardy-Weinberg平衡(P>0.05)。POU1F1:g.34236170A>G位点AA基因型个体第1胎断奶重显著高于GG基因型个体(P<0.05);FOLR1:g.31625869T>G位点TT基因型个体第1胎断奶重显著高于TG基因型个体(P<0.05),TT基因型个体第2胎和3胎联合断奶重显著高于TG基因型和GG基因型个体(P<0.05);FOLR1:g.31626833G>A位点AA基因型个体3胎联合断奶重显著高于GG基因型个体(P<0.05)。说明POU1F1和FOLR1基因的SNPs位点与努比亚山羊的断奶重相关,可作为山羊品种繁殖性状选育的候选基因和遗传分子标记。To explore the association of polymorphism in POU1 F1 and FOLR1 genes with weaning weight in Nubian goats, in this study, 355 Nubian goats were selected as research subjects. DNA mixing pool and Sanger sequencing were used to detect the candidate sites of these two genes. The SNPs sites were genotyped by MTA-seq sequencing technology. Hardy-Weinberg equilibrium was determined and population genetic parameters were calculated according to typing statistics. Moreover, GLM model was used to analyze the correlation between gene polymorphism and weaning weight of Nubian goats. The results showed that a polymorphic site POU1 F1:g.34236170 A>G was detected in POU1 F1 gene, including AA, AG and GG genotypes, and AA was the dominant genotype and A was the dominant allele. Two SNPs sites, FOLR1:g.31625869 T>G and FOLR1:g.31626833 G>A, were detected in FOLR1 gene. FOLR1:g.31625869 T>G site contains TT, TG and GG genotypes, with GG as the dominant genotype and G as the dominant allele. FOLR1:g.31626833 G>A site contains GG, GA and AA genotypes, with GG as the dominant genotype and G was the dominant allele. POU1 F1:g.34236170 A>G site and FOLR1:g.31625869 T>G site were moderately polymorphic(0.25A site was low and belonged to low polymorphic(PIC<0.25), and all the three sites conformed to Hardy-Weinberg equilibrium(P>0.05). The weaning weight of AA genotype at POU1 F1:g.34236170 A>G site was significantly higher than that of GG genotype in the first parity(P<0.05). The weaning weight of TT genotype at FOLR1:g.31625869 T>G site was significantly higher than that of TG genotype in the first parity(P<0.05). The weaning weight of TT genotype was significantly higher than that of TG and GG genotypes in the second and third parity(P<0.05). The weaning weight of AA genotype at FOLR1:g.31626833 G>A site was significantly higher than that of GG genotype in the total parity(P<0.05).
