摘 要:目的观察人参皂苷Rg3对重症急性胰腺炎(SAP)大鼠炎症介质的影响。方法取60只SD大鼠,随机取10只作为空白对照组,另50只腹腔注射雨蛙素联合脂多糖诱导建立重症急性胰腺炎模型后,随机分成SAP模型组、地塞米松组和高、中、低三个剂量的人参皂苷Rg3组;地塞米松组和人参皂苷Rg3组分别灌胃给予地塞米松或人参皂苷Rg3;建模24小时后处死大鼠,收集胰腺组织与血清,ELISA法检测血清IL-1β、TNF-α、IL-6的表达水平,干湿法检测胰腺水肿指数,血生化仪检测血清淀粉酶水平。结果与空白对照组比较,SAP模型组胰腺组织水肿指数、血清淀粉酶、IL-1β、TNF-α、IL-6表达水平显著增高(P<0.05);与SAP模型组比较,地塞米松组、不同剂量人参皂苷Rg3干预组胰腺组织水肿指数、血清淀粉酶、IL-1β、TNF-α、IL-6表达水平显著降低(P<0.05)。结论人参皂苷Rg3可通过抑制炎症因子减轻SAP大鼠的炎症反应。Objective To investigate the protective effect of ginsenoside Rg3 on rates with severe acute pancreatitis.MethodsSixty SD rats were selected and 10 rats were randomly selected as the blank control group;and after the severe acute pancreatitis model was established by intraperitoneal injection of cerulein combined with lipopolysaccharide,50 rats were randomly divided into the SAP model group,the dexamethasone group and ginsenoside Rg3 groups with high,medium and low doses;the dexamethasone group and ginsenoside Rg3 groups were given dexamethasone and Ginsenoside Rg3 by intragastric gavage,respectively.The rats were sacrificed after 24 hours,pancreatic tissues and serum were collected,ELISA was used to detect the expression levels of IL-1β,TNF-αand IL-6,dry-wet method was used to detect pancreatic edema index,and blood biochemical analyzer was used to detect serum amylase level.Results Compared with those in the blank control group,the pancreatic tissue edema index,the serum amylase,and the expression levels of IL-1β,TNF-αand IL-6 significantly increased in the SAP model group(P<0.05).Compared with those in SAP model group,the pancreatic tissue edema index,the serum amylase,and the expression levels of IL-1β,TNF-αand IL-6 significantly decreased in the dexamethasone group and the Ginsenoside Rg3 intervention groups(P<0.05).Conclusion Ginsenoside Rg3 can alleviate severe acute pancreatitis in rats by inhibiting inflammation.
