摘 要:目的:研究不同的超滤液用量对C群脑膜炎奈瑟球菌精制多糖质量的影响。方法:对C群脑膜炎奈瑟球菌多糖苯酚法去蛋白后的多糖液进行超滤,超滤液为0.5 mol/L氯化钙溶液,反复浓缩超滤。当超滤液用量达到20 L、30 L、40 L和50 L时,分别从截留管处取多糖液5 mL用于苯酚残留量的检测。对比超滤液用量为20 L和50 L时精制多糖的分子量、回收率、细菌内毒素和收率结果。结果:随着超滤液用量的增大,苯酚残留量逐渐降低;超滤液用量为50 L时,多糖分子量升高,精制多糖收率略有提高;细菌内毒素没有明显变化。结论:当超滤液用量为50 L时,多糖液中苯酚残留量降为0 g/L,多糖分子量升高,精制多糖收率略有增加,从而提高了多糖产品的质量。Objective:To study the effect of different dosage of ultrafiltrate on the quality of polysaccharide refined from Neisseria meningitidis Group C.Methods:After protein removal by phenol method,the polysaccharide solution of Neisseria meningitidis group C is ultrafiltration.The ultrafiltrate is 0.5 mol/L calcium chloride solution.The polysaccharide solution is concentrated by ultrafiltration repeatedly.When the amount of ultrafiltrate reached 20 L,30 L,40 L and 50 L,5 mL of polysaccharide solution is taken from the interception tube for the detection of phenol residue respectively.The results of polysaccharide molecular weight,polysaccharide recovery,bacterial endotoxin and yield of the refined polysaccharides are compared when the dosage of ultrafiltrate is 20 L and 50 L.Results:With the increase of the amount of ultrafiltrate,the residual amount of phenol decreased gradually.When the dosage of ultrafiltrate is 50 L,the molecular weight and the recovery rate of polysaccharide increased.The yield of refined polysaccharide increased slightly.There is no significant change in bacterial endotoxin.Conclusion:When the dosage of ultrafiltrate reached 50 L,the residual amount of phenol in the polysaccharide solution is reduced to 0 g/L,the molecular weight of polysaccharide is increased,and the yield of refined polysaccharide is slightly increased,thus improving the quality of polysaccharide products.
