摘　　要：本工作研制了一种细胞荧光成像探针,它可以快速高效地识别靶标细胞。探针由四种寡聚核苷酸片段所杂交形成,核心部分是一个十字型DNA结构,包含四个黏性末端、一个核酸适体端头,一个生物素端头。其中核酸适体赋予探针靶向功能,以便探针能够定位到目标细胞,生物素端头可连接荧光量子点,粘性末端使得十字型DNA自组装成为纳米级的探针颗粒。实验展示了该探针在加标血样中出色的染色效果,展现了良好的特异性和较强的荧光强度。该方法可用于复杂混合细胞样品的体外荧光成像检测。In this work,we developed a fluorescent cell imaging probe,which can quickly and efficiently identify target cells.The probe is formed by hybridization of four oligonucleotide fragments.The core part of the probe is a cross-shaped DNA structure consisting of four sticky ends,an aptamer end,and a biotin end.Among them,nucleic acid aptamer gives the probe targeting function,so that the probe can locate the target cell,the biotin terminal can be connected with fluorescent quantum dots,and the sticky terminal makes the cross-shaped DNA self-assemble into nanoscale probe particles.The results show that the probe has excellent staining effect in blood samples with added standard,showing good specificity and strong fluorescence intensity.This method can be used for fluorescence imaging detection of complex mixed cell samples in vitro.
