摘 要:目的探讨匹诺塞林对缺糖缺氧/复糖复氧(OGD/R)损伤SH-SY5Y细胞的细胞内钙离子([Ca^(2+)]i)变化及Caspase-12表达的影响。方法SH-SY5Y细胞制备OGD/R损伤模型,缺糖缺氧2 h,复糖复氧12 h,荧光探针法检测细胞胞质内钙信号变化,免疫荧光检测细胞内Caspase-12表达。结果OGD/R损伤可使SH-SY5Y[Ca^(2+)]i升高,Caspase-12表达增加,匹诺塞林能降低OGD/R损伤的SH-SY5Y细胞的[Ca^(2+)]i,减少Caspase-12的表达。结论匹诺塞林能减轻OGD/R诱导的SH-SY5Y细胞损伤,作用机制可能与其降低[Ca^(2+)]i,减少Caspase-12表达有关。Objective To discuss the effects of pinocembrin on intracellular calcium ion([Ca^(2+)]i)and the expression of Caspase-12 in SH-SY5Y cells injured by oxygen-glucose deprivation/reperfusion(OGD/R).Methods SH-SY5Y cells were used to establish OGD/R injury model with oxygen-glucose deprivation for 2 h and reperfusion for 12 h,then the intracellular calcium signal was detected by fluorescent probe,and the expression of intracellular Caspase-12 was detected by immunofluorescence.Results OGD/R injury can increase SH-SY5Y[Ca^(2+)]i and increase the expression of Caspase-12.Pinocerin can reduce[Ca^(2+)]i and reduce the expression of Caspase-12 in SH-SY5Y cells injured by OGD/R.Conclusion Pinocerin can relieve the injury of SH-SY5Y cells induced by OGD/R,and the mechanism of action may be related to the reduction of[Ca^(2+)]i and the expression of Caspase-12.
