龟甲胶对环磷酰胺引起的贫血大鼠的改善作用研究

摘　　要：目的:观察龟甲胶对环磷酰胺引起的贫血大鼠的改善作用。方法:将所有大鼠随机分为空白组、模型组、利可君组、龟甲胶低剂量组、龟甲胶中剂量组和龟甲胶高剂量组,每组12只,雌雄各半。除空白组外,均腹腔注射环磷酰胺建立贫血模型。造模成功后,空白组和模型组给予生理盐水,其余各组给予相应药物,连续灌胃15 d,最后一次灌胃24 h后处死。于腹主动脉采血,用自动血细胞分析仪检测外周血象,ELISA法检测血清EPO含量,RT-qPCR法检测肾脏EPO mRNA表达。结果:与模型组比较,龟甲胶组大鼠血液中WBC、RBC、HGB显著升高(P<0.01),血清中EPO含量显著升高(P<0.01);肾脏中EPO mRNA表达均显著上升(P<0.05)。结论:龟甲胶能够改善环磷酰胺引起贫血的大鼠WBC、RBC、HGB含量,并且促进肾脏中EPO mRNA表达。Objective:To observe the improvement effect of tortoise shell glue on CTX-induced anemia in rats.Methods:72 SD rats were randomly divided into the blank group,the model group,the Leucogen Tablets group,the tortoise shell glue groups of low-dose,medium-dose and high-dose,with 12 rats in each group.Anemia model was established by CTX injection.After successful modeling,corresponding interventions were given in each group for 15 days.24 hours after the last administration,blood was collected from the abdominal aorta,and the peripheral blood was detected with an automatic blood cell analyzer;the serum content of EPO was detected by ELISA,and renal EPO mRNA expression was detected by RT-qPCR.Results:Compared to those in the model group,the contents of WBC,RBC and HGB were significantly increased(P<0.01),the serum content of EPO was significantly increased(P<0.01),and the renal EPO mRNA expression was also greatly increased in the tortoise shell glue groups(P<0.05).Conclusion:Tortoise shell glue can improve the contents of WBC,RBC and HGB and promote the renal EPO mRNA expression in rats with anemia caused by CTX.
