摘 要:为调查贝氏隐孢子虫单独以及与禽致病性大肠杆菌合并感染对雏鸡肠道菌群的影响,将1日龄SPF级雏鸡分为隐孢子虫单独感染组(灌胃隐孢子虫1×10^(6)个/只)、隐孢子虫和大肠杆菌共感染组(肌注大肠杆菌2×106 CFU/只,同时灌胃隐孢子虫1×10^(6)个/只)与健康对照组,分别于接种后第3、9、15、21和27天采集新鲜粪便,提取DNA后进行16S rRNA高通量测序及生物信息学分析。结果显示,3组雏鸡肠道微生物相对丰度最高的门为厚壁菌门,其次为拟杆菌门和变形菌门。多样性分析表明,2组感染雏鸡的肠道菌群多样性降低,但隐孢子虫单独感染组较共感染组先恢复。从属水平分析发现,与健康对照组比较,共感染组的乳杆菌属丰度明显降低(P<0.05),而大肠/志贺菌属则显著升高(P<0.05),两者此消彼长。2感染组雏鸡肠道中的布劳特菌属和瘤胃球菌属伴随雏鸡日龄增长呈上升趋势,但相比健康对照组绝对丰度较低。以上结果表明,大肠杆菌的共感染可加剧隐孢子虫感染造成的雏鸡肠道微生物多样性和菌群结构的变化,这可能与采食量的改变以及对免疫力和代谢功能的影响有关。To understand the impact of Cryptosporidium bailei alone and in combination with avian pathogenic Escherichia coli on the intestinal flora of chicks after infection,1-day-old SPF chicks were used and divided into 3 groups:the groups infected with C.baileyi(by intragastric administration),the group infected with E.coli(by intragastric administration)combined with DE17(by intramuscular injection),and the healthy control group(without infection).On the 3 rd,9 th,15 th,21 st and 27 th days after infection,the fresh feces were collected from the chickens,and 16S rRNA high-throughput sequencing was used for bioinformatics analysis of the samples.The results showed that the phylum with the highest relative abundance of microorganisms in the three groups of chicks was Firmicutes,followed by Proteobacteria and Bacteroidetes.Diversity analysis showed that the diversity of the intestinal flora of the two groups of infected chicks decreased,but the C.baileyi infected group recovered earlier than the C.baileyi and E.coli co-infected group.The subordinate level analysis showed that,compared with the healthy control group,the abundance of Lactobacillus in the C.baileyi and E.coli co-infected group was significantly reduced(P<0.05),while their Escherichia-Shigella was significantly increased(P<0.05).The changes in the above flora caused significant changes in the risk of cardiovascular disease and xenobiotics,biodegradation and metabolism in the infected groups.The Blautella and Rumenococcus in the intestines of the infected chicks in the two groups showed an upward trend with the increase of the age of the chicks,but their absolute abundance was lower than that in the healthy control group.The above results indicate that the co-infection of C.baileyi and E.coli exacerbated the changes in the microbial diversity and microbial structure of the chicks caused by Cryptosporidium infection,which might be related to the changes in feed intake and the impact on immunity and metabolic functions in the chicks.
