江苏地区鸡传染性支气管炎核酸检测及其病毒分离株鉴定和基因序列分析

摘　　要：为了解江苏省蛋鸡传染性支气管炎病毒(infectious bronchitis virus,IBV)的主要流行情况,采集江苏省13个地级市20家蛋鸡养殖场鸡群的咽拭子及泄殖腔拭子样品共计625份。除去核酸浓度低于检出下限的120份样品后,通过RT-PCR的方法对余下的505份合格样品进行流行病学检测,并通过病料接种SPF鸡胚分离到1株IBV病毒,对该分离株进行全基因测序及S1基因序列分析。结果显示:505份合格样品中,IBV阳性检出率为14.46%(73/505)。IBV分离株全基因组大小为27689 bp,其中S1基因1620 bp,编码540个氨基酸,裂解位点为HRRRR,属于GⅠ-19谱系(QX型),分离株S1基因的核苷酸和氨基酸序列与GⅠ-19谱系(QX型)参考毒株同源性最高,分别为94%和95.4%。研究结果初步明确了江苏分离株遗传演化特点,为下一步防控工作提供了参考。To investigate the epidemiology of the infectious bronchitis virus(IBV)in chickens in Jiangsu Province in 2020,we collected 625 epidemic swab specimens on the chicken farms inthe 13 regions of Jiangsu Province.120 of the samples were unqualified whose nucleic acid concentration was lower than the detection limit,and the remaining 505 qualified samples were tested for epidemiology by RT-PCR.Then,we isolated an IBV virus by inoculating the pathogenic materials into SPF embryos and detected the full genomic sequence of the virus.The results were that the positive rate of IBV of the samples was 14.46%(73/505),and that the complete genome size of the isolated IBV strain was 27689 bp.Further analyzation showed that the S1 gene of the IBV was 1620 bp in length,and it encode 540 amino acids whit a cleavage site of HRRRR.Phylogenetic analysis revealed that the isolated strain was clustered into the genetic group GI-19(QX-like type).Homology analysis demonstrated that the nucleotide and deduced amino acid sequence of the S1 gene of the isolated strain were highly homologous with the reference strain of the same genotypes at 94%and 95.4%,respectively.In this study,the genetic evolution characteristics of the IBV isolates in Jiangsu Provincewere identified,which provided reference for further prevention and control of the virus.
