摘 要:目的以甘肃境内不同产地罂粟为研究对象,测定其不同部位(根、茎、叶、壳)中吗啡、可待因、蒂巴因、罂粟碱、那可丁5种生物碱的含量,为其合理开发利用和科学监管提供参考。方法采用高效液相色谱法,色谱柱为XDB-C18柱(250 mm×4.6 mm,5μm),以甲醇(A)-庚烷磺酸钠溶液(B)(称取庚烷磺酸钠1.01 g,加水适量使溶解,混匀,用水稀释至1000 ml,用磷酸调节pH值至2.6)为流动相,梯度洗脱(0~2 min,15%A;2~35 min,15%~40%A;35~80 min,40%A);流速为1.0 ml/min;检测波长:238 nm(罂粟碱),216 nm(其余4个生物碱);柱温:20℃;进样量:10μl。结果吗啡、可待因、蒂巴因、罂粟碱、那可丁分别在10.12~101.2,10.17~101.7,1.535~30.70,2.362~47.24,2.314~57.85μg/ml浓度范围内呈良好线性关系(r>0.9980);平均加样回收率为91.04%~99.44%,RSD为1.07%~1.59%(n=6);罂粟壳中吗啡、可待因、蒂巴因、罂粟碱、那可丁的含量为2.617~3.582,0.4859~0.8747,0.0665~0.0994,0.3221~0.5537,0.4095~0.5806 mg/g;罂粟根、茎、叶中吗啡含量为0.924~1.351,0.314~0.458,0.984~1.421 mg/g,可待因含量为0.5414~0.8305,0.2147~0.3214,0.4765~0.6247 mg/g,蒂巴因、罂粟碱、那可丁未检出。结论此法简便可行、灵敏准确,可为其合理开发利用和科学监管提供参考。Objective To determine morphine,codeine,thebaine,papaverine and narcotine in different parts(roots,stems,leaves and shell)of Papaver somniferum L.from different origins in Gansu Province to provide reference for its rational development,utilization and scientific supervision.Methods HPLC analysis was performed on a XDB-C_(18) column(250 mm×4.6 mm,5μm).The mobile phase consisted of methanol(A)-sodium heptane sulfonate solution(B)(weigh 1.01 g of sodium 1-heptanesulfonate,add appropriate amount of water to dissolve it,mix it evenly,dilute it with water to 1000 ml,and adjust the pH value to 2.6 with phosphoric acid),and gradient elution was performed(0-2 min,15%A;2-35 min,15%A-40%A;35-80 min,40%A).The flow rate was 1 ml/min.The detection wavelengths were 238 nm for papaverine and 216 nm for the others.The column temperature was 20℃and the injection volume was 10μl.Results The contents of morphine,codeine,thebaine,papaverine and narcotine had good linear relationships with the peak areas in the ranges of 10.12-101.2,10.17-101.7,1.535-30.70,2.362-47.24 and 2.314-57.85μg/ml(r>0.9980),respectively.The average recoveries were 91.04%-99.44%,with RSDs of 1.07%-1.59%(n=6).The contents of morphine,codeine,thebaine,papaverine and narcotine in the shells were 2.617-3.582,0.4859-0.8747,0.0665-0.0994,0.3221-0.5537 and 0.4095-0.5806 mg/g,respectively.The contents of morphine in the roots,stems and leaves were 0.924-1.351,0.314-0.458 and 0.984-1.421 mg/g,respectively,and the contents of codeine were 0.5414-0.8305,0.2147-0.3214 and 0.4765-0.6247 mg/g,respectively,but thebaine,papaverine and narcotine were not detected.Conclusion This method is simple,sensitive and accurate,and can provide reference for reasonable utilization and scientific supervision on Papaver somniferum L.
