摘 要:目的建立高效液相色谱法同时测定正柴胡饮颗粒中柴胡皂苷a、柴胡皂苷d、橙皮苷、升麻素苷、5-O-甲基维斯阿米醇苷、甘草苷、甘草酸铵、甘草次酸和芍药苷的含量。方法应用HPLC-DAD多波长切换梯度洗脱技术,采用Agilent TC-C18色谱柱(250 mm×4.6 mm,5μm),以乙腈(A)-0.02%甲酸水溶液(B)为流动相,流速1.0 ml/min;检测波长为283 nm(橙皮苷)、230 nm(芍药苷、甘草苷)、254 nm(升麻素苷、甘草酸铵、甘草次酸、5-O-甲基维斯阿米醇苷)、210 nm(柴胡皂苷a、柴胡皂苷d);柱温为25℃,进样量为10μl。结果橙皮苷、芍药苷、甘草苷、升麻素苷、甘草酸铵、甘草次酸、柴胡皂苷a、柴胡皂苷d和5-O-甲基维斯阿米醇苷质量浓度的线性范围分别为2.111~42.220,4.657~93.140,2.883~57.660,1.487~29.740,1.944~38.880,0.907~18.140,1.235~24.700,0.817~16.340,0.844~16.880μg/ml(r为0.9994~0.9999);平均加样回收率分别为99.2%,101.0%,100.2%,99.7%,99.8%,98.3%,98.7%,101.6%,101.0%(RSD<2.0%,n=6);精密度、重复性、稳定性(24 h)试验的RSD<2.0%(n=6)。结论该方法可用于正柴胡饮颗粒的多指标成分的质量控制研究。Objective To establish a method for simultaneous determination saikosaponin a,saikosaponin d,hesperidin,prim-O-glucosylcimifugin,5-O-methylvisammioside,liquiritin,ammonium glycyrrhizinate,clycyrrhetinic acid and paeoniflorin in Zhengchaihuyin Granules by HPLC.Methods The detection was carried out on an Agilent TC-C_(18)(250 mm×4.6 mm,5μm)column with the mobile phase consisted of acetonitrile(A)-0.02%formic acid aqueous solution(B)in a gradient mode at a flow rate of 1.0 ml/min.The detection wavelength was set at 283 nm for hesperidin,230 nm for paeoniflorin and liquiritin,254 nm for prim-O-glucosylcimifugin,ammonium glycyrrhizinate,clycyrrhetinic acid and 5-O-methylvisammioside,210 nm for saikosaponin a and saikosaponin d.The column temperature was set at 25℃and the injection volume was 10μl.Results The linear range was 2.111-42.220μg/ml for hesperidin,4.657-93.140μg/ml for paeoniflorin,2.883-57.660μg/ml for liquiritin,1.487-29.740μg/ml for prim-O-glucosylcimifugin,1.944-38.880μg/ml for ammonium glycyrrhizinate,0.907-18.140μg/ml for clycyrrhetinic acid,1.235-24.700μg/ml for saikosaponin a,0.817-16.340μg/ml for saikosaponin d,0.844-16.880μg/ml for 5-O-methylvisammioside(r=0.9994-0.9999);the average recoveries were 99.2%,101.0%,100.2%,99.7%,99.8%,98.3%,98.7%,101.6%,101.0%(RSD<2.0%,n=6).RSDs of precision,reproducibility and stability tests(24 h)were lower than 2.0%(n=6).Conclusion The method can be used for quality control of multicomponents in Zhengchaihuyin Granules.
