痔脱垂组织中TNF-α、IL-1β、IFN-γ表达和Treitz肌形态及密度变化的研究

摘　　要：目的:探讨痔脱垂组织中TNF-α、IL-1β、IFN-γ表达和Treitz肌形态及密度变化。方法:选取2010年5月至2014年4月20例脱垂性痔组织(Ⅲ和Ⅳ度)为实验组,20例低位直肠癌患者正常痔组织为对照组,采用HE染色、免疫组织化学染色技术及光密度测定方法,对比两组痔组织中TN F-α、IL-1β、IFN-γ表达和Treitz肌的形态及密度变化。结果:实验组、对照组痔组织TNF-α、IL-1β和IFN-γ平均光密度值分别为(107.98±32.47)、(38.80±5.30),(99.87±24.17)、(34.80±5.61),(97.82±30.47)、(32.76±1.93),两组比较差异具有统计学意义(P<0.05)。实验组痔组织Treitz肌疏松,出现Treitz肌断裂、扭曲、变形及玻璃样变等变化;对照组痔组织Treitz肌密集,形态较规则,断裂和变形少见。实验组、对照组痔组织Treitz肌平均光密度值分别为13.40±5.10、85.41±34.47,两组比较差异有统计学意义(P<0.01)。结论:脱垂性痔组织中TNF-α、IL-1β和IFN-γ表达增加,Treitz肌形态发生改变、光密度值明显降低,为探讨痔发生发展的规律,制定更加准确的临床诊治措施提供了依据。Objective:To investigate the expression of TNF-α,IL-1β,IFN-γand the changes of Treitz muscle morphology and density in hemorrhoids prolapse tissue.Methods:From May 2010 to April 2014,20 cases of prolapsed hemorrhoids(III and IV degree hemorrhoids)were selected as the experimental group and 20 cases of normal hemorrhoids in patients with low rectal cancer as the control group.The expression of TN F-α,IL-1βand IFN-γ,the morphology and density of Treitz muscle in hemorrhoids of the two groups were compared and analyzed by HE stain,immunohistochemical technology staining and optical density measurement.Results:The average optical density values of TNF-α,IL-1βand IFN-γin hemorrhoids of experimental group and control group were(107.98±32.47),(38.8±5.3),(99.87±24.17),(34.80±5.61),(97.82±30.47),(32.76±1.93),respectively,with statistical significance(P<0.05).In the experimental group,the Treitz muscle was loose,and the Treitz muscle was broken,twisted,deformed and glassy.In the control group,the Treitz muscle was dense,regular,and rarely broken or deformed.The average optical density of Treitz muscle in experimental group and control group was 13.40±5.10 and 85.41±34.47,respectively,with significant difference(P<0.01).Conclusion:The expression of TNF-α,IL-1βand IFN-γin prolapsed hemorrhoids increased,the morphology of Treitz muscle changed,and the optical density decreased significantly.This study provides a basis for exploring the law of the development of the occurrence of hemorrhoids and formulating more accurate clinical diagnosis and treatment measures.
