摘 要:目的探讨ICSⅡ干预自发性高血压大鼠(Spontaneously hypertensive rats,SHR)左心室重构是否与其上调PGC-1α,促进心肌脂肪酸β-氧化有关。方法将14周龄雄性SHR随机均分为模型组(SHR组)和ICSⅡ干预组(ICSⅡ组,8 mg/kg),同时以同龄雄性魏-凯二氏大鼠(Wistar Kyotorats,WKY)作为空白对照组。ICSⅡ组每日灌胃1次,SHR和WKY组给予等体积双蒸水。给药12周后称量大鼠体重、全心重,计算心脏质量指数。H&E染色观察大鼠左心室病理变化。Western blot检测左心室组织PGC-1α、CPT-1α、MCAD的蛋白表达。结果与WKY组相比,SHR组大鼠心脏质量指数明显升高(P<0.05);左心室心肌细胞肥大、排列紊乱、炎性细胞浸润;左心室PGC-1α、CPT-1α、MCAD的蛋白表达均降低(P<0.05)。与SHR组相比,ICSⅡ组大鼠心脏质量指数明显降低(P<0.05);左心室病理变化明显改善;左心室PGC-1α、CPT-1α、MCAD的蛋白表达均升高(P<0.05)。结论ICSⅡ至少可能通过上调PGC-1α、促进心肌脂肪酸β-氧化,而改善SHR左心室重构。Objective To investigate whether IcarisideⅡ(ICSⅡ)improved left ventricular remodeling in male spontaneously hypertensive rats(SHR)through up-regulating PGC-1αand promoting myocardial fatty acidβ-oxidation.Methods The 14-week age SHR were randomly divided into SHR group and ICSⅡ(8 mg/kg)-treatment group.Male Wistar Kyoto rats(WKY)of 14-week age were set as control group.The ICSⅡgroup was given intragastrically once a day.The SHR and WKY groups were given the match volume of double distilled water.After 12 weeks of administration,the body weight and heart weight were weighed separately and the heart mass index was calculated.The pathological changes of left ventricle were observed by H&E staining.The protein expressions of PGC-1α,CPT-1αand MCAD in the left ventricle were measured by Western blot assay.Results Compared with the WKY group,the heart mass index was increased,and the changes of myocardial hypertrophy,myofibrillar disorder and inflammatory cells infiltration were observed.The protein expressions of PGC-1α,CPT-1αand MCAD were down-regulated in SHR group.Compared with the SHR group,the heart mass index was decreased,and the pathological changes of left ventricle were improved and the protein expressions of PGC-1α,CPT-1αand MCAD were up-regulated in the ICSⅡgroup.Conclusion ICSⅡimproves the left ventricular remodeling in SHR,and the mechanism might be at least partially through up-regulating PGC-1αand promoting myocardial fatty acidβ-oxidation.
