摘 要:为研究海螵蛸抗菌活性,本文通过比较海螵蛸的不同处理方法,采用药敏纸片扩散法和二倍稀释法测定了金乌贼(Sepia esculenta)、曼氏无针乌贼(Sepiella maindroni)、虎斑乌贼(Sepia pharaonis)3种海螵蛸对金黄色葡萄球菌(Staphylococcus aureus)、铜绿假单胞菌(Pseudomonas aeruginosa)、哈氏弧菌(Vibrio harveyi)等8种人体及海水常见菌的抑菌效果。结果显示:海螵蛸粉末用80%酒精、按照固液比1∶4,80 W超声波3次,60℃,每次20 min震荡6 h混合后抗菌活性最好。3种海螵蛸均对哈氏弧菌(V.harveyi)、溶藻弧菌(Vibrio alginolyticus)、金黄色葡萄球菌(S.aureus)抑制效果较好,其中,对哈氏弧菌(V.harveyi)抑制效果最强,溶藻弧菌(V.alginolyticus)其次,金黄色葡萄球菌(S.aureus)居于第三。3种海螵蛸中,金乌贼(S.esculenta)和曼氏无针乌贼(S.maindroni)海螵蛸抑制效果相似且强于虎斑乌贼(S.pharaonis)。在8种菌中,对抑制效果最明显的哈氏弧菌(V.harveyi),最小抑菌浓度(Minimum inhibitory concentration,MIC)为3.9 mg/mL,其次是溶藻弧菌(V.alginolyticus),MIC为15.6 mg/mL。研究结果表明三种海螵蛸均具有抗菌活性,这为海螵鞘应用价值提供了理论依据。To study the antibacterial activity of cuttlebone in vitro,Sepia esculenta cuttlebone was used to determine the optimal liquid treatment method.The antibacterial activity of three cuttlebone species(S.esculenta,Sepiella maindroni,Sepia pharaonis)against 8 common human and seawater bacteria including Staphylococcus aureus,Pseudomonas aeruginosa,and Vibrio harveyi was determined by susceptibility disk diffusion method and double dilution method in vitro.The results showed that the ultrasonic treatment group had the best effect in different sterilization treatment,ultraviolet irradiation,high pressure soaking at 121℃and boiling had no antibacterial effect;80%ethanol had the best effect in the five solvents,the next is 60%ethanol,the distilled water and 0.9%NaCl solution had the worst effect;the best effect of different solid solution ratios was 1∶4,and 1∶5 had the worst effect;and 6 h is the best time to soak cuttlebone.Therefore,in the detection of antibacterial activity,cuttlebone powder was detected after they mixed with 80%alcohol,1∶4 ratio of solid to liquid,80 W ultrasound for 3 times,60℃,20 min each time,and shook for 6 hours.All three cuttlebone showed good inhibitory effects on V.harveyi,V.alginolyticus,and S.aureus,and the inhibitory effects were V.harveyi>V.alginolyticus>S.aureus.Among the cuttlebones of three species,the inhibitory effects of S.esculenta and S.maindroni cuttlebone were similar and stronger than S.pharaonis cuttlebone,and among the eight bacteria,S.esculenta and S.maindroni cuttlebone had the best inhibitory effect on V.harveyi,with a minimum inhibitory concentration(MIC)of 3.9 mg/mL,followed by V.alginolyticus,with an MIC of 15.6 mg/mL.The results showed that all three cuttlebone had antibacterial activity and could be used as by-products of seafood processing.
