摘 要:目的探讨勿动蛋白(Nogo)与Rho相关蛋白激酶(ROCK)对心脏交感神经重塑与去甲肾上腺素转运蛋白表达的调节作用。方法建立SD乳鼠脊髓背根神经元细胞与心肌细胞联合培养的模型,随机数表法分为A组(心肌细胞+交感神经节细胞);B组(心肌细胞+交感神经节细胞+Nogo-66);C组(心肌细胞+交感神经节细胞+C3转移酶+Nogo-66);D组(心肌细胞+交感神经节细胞+Y-2763+Nogo-66)。倒置相差显微镜观察神经元细胞与心肌细胞共培养细胞的生长状况。免疫荧光测定酪氨酸羟化酶(TH)与去甲肾上腺素转运蛋白表达。ELISA检测培养液中去甲肾上腺素含量(NE)。结果免疫荧光染色结果显示,与A组(1.00±0.00)比较,B组(0.07±0.01)TH表达明显降低,C组(1.99±0.55)与D组(2.13±0.72)TH表达明显增加,差异均有统计学意义(P<0.05)。ELISA检测结果显示,与A组(117.34±2.47)ng/L比较,B组[(94.32±7.34)ng/L]去甲肾上腺素含量明显降低,C组[(124.61±2.97)ng/L)与D组[(127.19±3.54)ng/L)去甲肾上腺素含量明显增加,差异有统计学意义(P<0.05)。免疫荧光染色结果显示,与A组(1.00±0.00)比较,B组(0.07±0.01)TH表达明显降低,C组(1.99±0.55)与D组(2.13±0.72)TH表达明显增加,差异有统计学意义(P<0.05)。。结论Nogo与ROCK参与心脏交感神经重塑与去甲肾上腺素转运蛋白表达的调节。Objective To investigate cardiac sympathetic nerve remodeling and expression of norepinephrine transporter(NET)regulated by Nogo and ROCK.Methods Thirty SD rats aged 1-3 days were used to prepare cardiomyocytes and spinal dorsal root neurons,and establish a model of co-culture of spinal dorsal root neurons and cardiomyocytes.The cell model was randomly divided into four groups:group A:cardiomyocytes and sympathetic ganglion cells;group B:cardiomyocytes,sympathetic ganglion cells and Nogo-66;group C:cardiomyocytes,sympathetic ganglion cells,C3 transferase and Nogo-66;group D:cardiomyocytes,sympathetic ganglion cells,Y-2763 and Nogo-66.Expression of tyrosine hydroxylase(TH)and norepinephrine transporter was detected by immunofluorescence.Norepinephrine(NE)was detected by ELISA.Results Compared with group A(1.00±0.00),expression of TH in group B(0.07±0.01,P=0.035)was significantly lower than that in group A(1.99±0.55,P=0.027)and D(2.13±0.72,P=0.015).Compared with group A(117.34±2.47)ng/L,the norepinephrine content in group B[(94.32±7.34)ng/L,P<0.001]was significantly lower than that in group A(124.61±2.97)ng/L,P=0.021)and D[(127.19±3.54)ng/L,P=0.003].Compared with group A(1.00±0.00),TH expression in group B(0.07±0.01,P=0.035)was significantly lower than that in group A(1.00±0.00),while those in group C(1.99±0.55,P=0.027)and group D(2.13±0.72,P=0.015)were significantly increased.Conclusion Nogo and ROCK are involved in regulation of cardiac sympathetic nerve remodeling and norepinephrine transporter expression.
