摘 要:为研究副猪嗜血杆菌(Haemophilus parasuis,HPS)相关sRNA(small RNA)的调控机制,以复苏培养后的2株HPS辽宁分离菌株为研究对象,通过实时荧光定量PCR对体外不同生长时期以及感染PK-15细胞后sRNA64和sRNA145的转录水平进行了分析。结果表明,体外生长过程中sRNA64、sRNA145分别在强、弱毒株中差异表达,2株HPS感染细胞后sRNA64均差异表达,而sRNA145仅在强毒株中差异表达。另外,sRNA64、sRNA145分别在强、弱毒株体外生长后期达到转录高峰,感染细胞后sRNA64在强、弱毒株感染早期达到转录高峰,而sRNA145仅在强毒株感染细胞后期达到转录高峰。说明sRNA64和sRNA145作为差异表达基因对调控HPS发挥着作用,研究结果为sRNA在不同毒力HPS致病机制中的进一步研究提供了基础数据。In order to study the regulation mechanism of small RNA(sRNA)related to Haemophilus parasuis,two H.parasuis strains isolated from Liaoning were used as the research objects.The transcription levels of sRNA64 and sRNA145 at different growth periods in vitro and after infection of PK-15 cells were analyzed by real-time quantitative PCR(RT-qPCR).The results showed that sRNA64 and sRNA145 were differentially expressed in the highly virulent strain and non-virulent strain during in vitro growth,and sRNA64 was differentially expressed after the two H.parasuis strains infected cells,while sRNA145 was only differentially expressed in the highly virulent strain.In addition,sRNA64 and sRNA145 reached the transcription peak in the late growth period of the highly virulent strain and non-virulent strain respectively.After infection of cells,sRNA64 reached the transcription peak in the early stage of infection of the highly virulent strain and non-virulent strain,while sRNA145 only reached the transcription peak in the late stage of the infection of the highly virulent strain.This shows that sRNA64 and sRNA145 as differentially expressed genes play a role in regulating H.parasuis,and this result provides basic data for further research on sRNA in the pathogenic mechanism of H.parasuis with different virulence.
