摘 要:为了解鸡喉黏膜不同时期的免疫状态,选择不同日龄海兰白鸡的喉组织,用免疫组织化学方法研究CD4^(+)、CD8^(+)T淋巴细胞亚群的出现、定位分布及数量变化过程。结果显示,4日龄时,CD4^(+)和CD8^(+)T淋巴细胞都首次出现在喉黏膜中,而且都主要分布在喉腔底壁的正中黏膜嵴中。7日龄时,喉黏膜中CD4^(+)和CD8^(+)T淋巴细胞的数量都显著增加,而且在喉黏膜中形成淋巴聚集物,CD4^(+)和CD8^(+)T淋巴细胞都主要分布在淋巴聚集物的中下部区域,而且主要以CD4亚群为主。14日龄和21日龄时CD4^(+)T淋巴细胞集中在黏膜固有层中下部区域,而CD8^(+)T淋巴细胞分散在整个黏膜固有层中,黏膜上皮内也有个别CD8^(+)T淋巴细胞存在。35日龄和56日龄时,CD4^(+)T淋巴细胞和CD8^(+)T淋巴细胞主要分布在生发中心周围的滤泡间区域,56日龄时形成喉黏膜相关淋巴组织。在数量变化上,随着日龄增长,CD4^(+)T淋巴细胞数量逐渐增加,而CD8^(+)T淋巴细胞从7日龄至35日龄,数量变化不大,而且显著低于CD4^(+)T淋巴细胞,但56日龄时CD8^(+)T淋巴细胞的数量迅速增加,并与CD4^(+)T淋巴细胞几乎相等。结果表明,鸡喉黏膜中T淋巴细胞亚群的分布和数量均呈现日龄相关性变化,并且在56日龄时形成典型的喉黏膜相关淋巴组织。In order to reveal the immune status of laryngeal mucosa of chickens at different stages,the larynx of Hy-Line white chickens at different days of age were selected in this study.The occurrence,location,distribution and number change of CD4^(+)and CD8^(+)T lymphocytes were investigated by immunohistochemical method.The results showed that both CD4^(+)and CD8^(+)T lymphocytes appeared in the laryngeal mucosa for the first time at 4-day-old,and they were mainly distributed in the midline mucosal crest at the bottom of laryngeal cavity.At 7-day-old,the number of CD4^(+)and CD8^(+)T lymphocytes increased significantly in the laryngeal mucosa,and lymphatic aggregates were formed in the laryngeal mucosa.CD4^(+)and CD8^(+)cells were mainly distributed in the middle and lower regions of the lymphatic aggregates,and the CD4 subsets were dominant.At 14 and 21-day-old,CD4^(+)cells were concentrated in the middle and lower lamina propria of mucosa,while CD8^(+)cells were scattered in the whole lamina propria of mucosa,and a few CD8^(+)cells also existed in the mucosa epithelium.At 35 and 56-day-old,CD4^(+)and CD8^(+)cells were mainly distributed in the interfollicular area around the germinal center,especially laryngeal mucosal associated lymphoid tissue(LALT)was formed at 56-day-old.The number of CD4^(+)T lymphocytes increased gradually with the increase of age,while the number of CD8^(+)T lymphocytes changed little from 7 to 35 day-old,and was significantly lower than that of CD4^(+)T lymphocytes.However,the number of CD8^(+)T lymphocytes increased rapidly and was almost equal to that of CD4^(+)T lymphocytes at 56-day-old.The results indicated that the distribution and number of T lymphocyte subsets in laryngeal mucosa of chickens showed age-related changes,and the typical LALT was formed at 56-day-old,and the tissue structure related to the immune function of laryngeal mucosa was mature.
