摘  要:目的探讨霍山石斛早期给药预防小鼠学习记忆能力减退的功效。方法33只雌性SPF级C57BL/6小鼠随机分为模型组、早期给药组和晚期给药组,另设12周龄小鼠为对照组,每组11只。采用Morris水迷宫检测学习记忆能力,透射电镜观察海马CA3区超微结构,免疫印迹检测海马CA3区SYT1表达。结果1~5 d逃避潜伏期,对照组分别为(50.33±14.07)s、(42.13±11.29)s、(28.24±17.78)s、(24.61±19.85)s、(16.11±6.18)s,模型组分别为(59.93±0.07)s、(52.37±6.47)s、(43.97±7.64)s、(42.34±15.10)s、(32.22±11.78)s,早期给药组分别为(53.81±13.59)s、(43.02±12.69)s、(29.40±7.79)s、(23.86±7.05)s、(22.96±8.45)s。靶象限游泳时间百分比,对照组、模型组、早期给药组和晚期给药组分别为(55.34±16.00)s、(40.70±9.97)s、(50.83±10.04)s、(41.62±10.03)s。与对照组比较,模型组逃避潜伏期1~5 d均延长,差异有统计学意义(F=10.539,P=0.000);靶象限游泳时间百分比减少,差异有统计学意义(P=0.020)。与模型组比较,早期给药组逃避潜伏期缩短2~5 d,靶象限游泳时间百分比增加,差异有统计学意义(P<0.05)。透射电镜显示,海马CA3区早期给药组和对照组相似,突触结构较清晰,紧靠突触前膜处具有完整结构的突触囊泡较多、分布均匀。与对照组比较,模型组海马CA3区SYT1高表达,差异有统计学意义(P<0.05);与模型组比较,早期给药组CA3区SYT1低表达,差异有统计学意义(P<0.05)。结论早期给予霍山石斛可以延缓或预防小鼠学习记忆能力减退,其机制可能与影响海马突触结构和突触蛋白SYT1的表达有关。Objective To elucidate the effect of early administration of Dendrobium huoshanense(DH) on preventing learning and memory impairment in mice and to explore its possible mechanism. Methods 33 SPF-grade C57 BL/6 mice were randomly divided into model group, early DH administration group and late DH administration group with 11 mice in each group.In addition, Eleven 12-week-old mice were included as the control group.Morris water maze was used to test the learning and memory ability of mice.Transmission electron microscopy was used to observe the synaptic ultrastructure of hippocampal CA3 region, and the expression of SYT1 in this region was determined by western blot. Results The escape latencies of 1-5 days were(50.33±14.07) s,(42.13±11.29) s,(28.24±17.78) s,(24.61±19.85) s and(16.11±6.18) s in the control group;(59.93±0.07) s,(52.37±6.47) s,(43.97±7.64) s,(42.34±15.10) s and(32.22±11.78) s in the model group;(53.81±13.59) s,(43.02±12.69) s,(29.40±7.79) s,(23.86±7.05) s and(22.96±8.45) s in the early DH administration group.The percentages of swimming time in target quadrant in control group, model group, early DH administration group and late administration group were(55.34±16.00) s,(40.70±9.97) s,(50.83±10.04) s and(41.62±10.03) s, respectively.Compared with the control group,the escape latencies of 1-5 days of mice in model group were all extended(F=10.539,P=0.000),while the percentages of target quadrant swimming time were shortened(P=0.020),the differences were statistically significant.Compared with the model group,the escape latencies of 2-5 days of mice in the early DH administration group were shortened,while the percentages of target quadrant swimming time were extended,the differences were also statistically significant(P<0.05).Transmission electron microscopy of hippocampal CA3 region showed a similar imaging between the early administration group and control group,with clear synaptic structure and more synaptic vesicles with complete structure evenly distributed in the place close to
