摘 要:目的拟改进PAS染色方法,以降低染色背景。方法取标本进行常规PAS染色,比较亮绿和苏木精衬染的染色效果,比较铬酸溶液浓度分别为5%、10%及20%的染色效果;采用10%浓度的铬酸溶液,比较Schiff溶液染色时间分别为15、30、60 min的染色效果;采用多种真菌感染性疾病的切片,包括体癣、脓癣以及深部真菌病行PAS-铬酸染色,验证改良PAS皮肤真菌染色的可靠性。结果常规PAS染色显示,亮绿衬染效果较好,且10%铬酸溶液氧化10 min浓度较佳;采用10%浓度的铬酸溶液,Schiff溶液染色时间为30 min最佳;体癣、脓癣以及深部真菌病的切片PAS-铬酸染色验证发现,该方案质量可靠。结论改良的PAS-铬酸染色方法比经典的PAS染色方法背景更干净,染色对比度较高。Objective To improve PAS staining method to reduce staining background.Methods Specimens were taken for routine PAS staining,and the staining effects of bright green and hematoxylin contrast staining were compared.The staining effects of chromate solution concentrations of 5%,10%and 20%were compared.Chromic acid solution with 10%concentration was used to compare the dyeing effects of Schiff solution at 15,30 and 60 min,respectively.PAS-chromate staining was performed on sections of various fungal infectious diseases,including tinea corporis,tinea pyogenes and deep fungal diseases to verify the reliability of modified PAS skin fungal staining.Results Conventional PAS staining showed that the effect of bright green was better,and the concentration of 10%chromic acid solution for 10 min was better.The best dyeing time was 30 min in 10%chromic acid solution.PAS-chromic acid staining of sections of tinea corporis,tinea pyogenes and deep fungal diseases showed that the scheme was reliable.Conclusion The modified PAS-chromic acid staining method has a cleaner background and higher staining contrast than the traditional PAS staining method.
