摘 要:目的探讨膝骨性关节炎(KOA)患者与健康对照者血清中白细胞介素15(IL-15)的变化。方法选取30例KOA患者和35例健康成人外周血,将KOA组分为两组,一是实验组加入IL-15(10 ng/ml),二是不加IL-15作为本组对照组。将HC组也分为两组,一是实验组加入IL-15(10 ng/ml),二是不加IL-15作为本组对照组。采用ELISA法检测KOA组和健康对照组外周血清中IL-15的水平,采用流式细胞仪检测IL-15对KOA患者和健康对照者外周血自然杀伤(NK)细胞受体表达的影响,以及采用ELISA法检测IL-15对外周血单个核细胞(PBMCs)产生肿瘤坏死因子α(TNF-α)和干扰素γ(IFN-γ)水平的影响。结果KOA组血清IL-15水平比对照组升高。IL-15可增强KOA组外周血NK细胞受体CD69、CD94和NKG2D的表达。KOA组细胞培养后上清液中TNF-α与IFN-γ的含量高于对照组。结论IL-15拮抗剂可能成为阻止或延缓KOA进展的一种新治疗选择。Objective To investigate the changes of serum interleukin-15(IL-15)in patients with knee osteoarthritis(KOA)and healthy controls.Methods The peripheral blood of 30 KOA patients and the peripheral blood of 35 healthy adults were selected.The KOA group was divided into two groups,one was the experimental group with IL-15(10 ng/ml),and the other was without IL-15 as the control group.The HC group was also divided into two groups,one was the experimental group with IL-15(10 ng/ml),and the other was without IL-15 as the control group.ELISA method was used to detect the level of IL-15 in peripheral blood of KOA group and healthy control group.Flow cytometry was used to detect the effect of IL-15 on the expression of natural killer(NK)cell receptor in peripheral blood of patients with KOA and healthy controls,and the ELISA method was used to detect the effect of IL-15 on the levels of tumor necrosis factorα(TNF-α)and interferonγ(IFN-γ)by peripheral blood mononuclear cells(PBMCs).Results The level of serum IL-15 in KOA group was higher than that in control group.IL-15 could enhance the expression of NK cell receptors CD69,CD94 and NKG2D in KOA group.The levels of TNF-αand IFN-γin the supernatant of KOA group were higher than those of control group.Conclusion IL-15 antagonist may become a new treatment option to prevent or delay the progression of KOA.
