摘  要:本研究旨在检测经气雾和口服途径接种布鲁氏菌疫苗(S2株)后羊特异性血清抗体阳性率的变化以确定其相对免疫功效。布鲁氏菌阴性羊群分别通过口服(1×1010 CFU剂量)和气雾(1×1010、8×109、6×109、4×109、2×109、1×109 CFU和5×108 CFU共7种剂量)接种布鲁氏菌疫苗(S2株)。接种后30、60、90、120 d和320 d分别采集试验羊的血清样品,并用虎红平板凝集试验(RBPT)和试管凝集试验(SAT)进行血清抗体检测。结果显示,口服免疫组接种后30d抗体阳性率达到最高(羔羊RBPT:38.00%,SAT:32.00%;成年羊RBPT:72.00%,SAT:68.00%),30~90 d抗体阳性率下降较快,90 d后抗体阳性率降到较低水平(RBPT﹤12.00%,SAT﹤10.00%);气雾免疫组,羔羊接种后30d,阳性率最高升至40.00%(RBPT:16.70%~40.00%,SAT:11.10%~30.00%),60d阳性率降至20.00%以内(RBPT:0~20.00%,SAT:5.60%~20.00%),90d阳性率降至10.00%以内,不同剂量气雾免疫羔羊阳性率变化差异不显著(P>0.05)。成年羊(除5×108 CFU组外),接种后30d阳性率升至84.81%(SAT:84.81%~88.50%),60d阳性率低于53.33%(SAT:44.30%~53.33%),90d阳性率低于24.39%(SAT:18.66%~24.39%),120d阳性率低于10.00%;5×108CFU组接种后30d阳性率较低(RBPT:48.70%,SAT:44.73%);成年羊气雾免疫组1×109~1×1010 CFU剂量同一时间点,阳性率结果变化差异不显著(P>0.05),与5×108 CFU组在30d和60d变化差异极显著(P<0.01)。口服免疫组抗体阳性率曲线变化趋势与气雾免疫组基本一致,接种后30d时都达到峰值,羔羊阳性率均较低,成年羊气雾免疫组阳性率高出口服免疫组18.84%。本试验表明,布鲁氏菌疫苗(S2株)气雾免疫在成年羊体内产生的特异性血清抗体阳性率优于口服免疫。This study aimed to examine positive rates of specific serum antibodies in sheep immunized with Brucella S2 vaccine by aerosol and oral routes and determine their relative immune efficiency.Experimental lambs and adult sheep were immunized by either oral route (dose 10 billion CFU) or aerosol route (doses 10,8,6,4,2,1,and 0.5 billion CFU).Serum samples were collected from immunized sheep on days 30,60,90,120,and 320 post immunization,and analyzed by rose bengal plate agglutination test (RBPT) and standard-tube agglutination test (SAT).The results showed that after oral immunization,positive rates of specific serum antibodies peaked on day 30 (lambs RBPT:38.00%,SAT:32.00%;adult sheep RBPT:72.00%,SAT:68.00%),the antibody positive rate decreased rapidly from 30 to 90 days,and then decreased to a low level by day 90 (RBPT<12.00%,SAT<10.00%).After aerosol immunization,positive rates of specific serum antibodies peaked at day 30 (lambs RBPT:16.70%-40.00%,SAT:11.10%-30.00%),then declined by days 60 and 90,with positive rates of 20.00%and 10.00%,respectively.Furthermore,positive rates of specific serum antibodies in lambs with aerosol immunization at different doses showed no significant differences (P>0.05);whereas positive rates in adult sheep were>84.81%(SAT:84.81%-88.50%) on day 30,<53.33%(SAT:44.30%-53.33%) on day 60,<24.39%(SAT:18.66%-24.39%) on day 90,and<10.00%on day 120,except for dose 5×108CFU group,which produced lower positive rate (RBPT:48.70,SAT:44.73%) on day 30.No significant difference in positive rate of immune dose from 1×109-1×1010CFU was observed at the same time point in aerosol immunization group (P>0.05),and the difference between immune dose of 5×108CFU group at 30d and 60d was significant (P<0.01).The variation trend of serum antibody positive rates in oral immunization group was basically the same as that in aerosol immunization group.On day 30,RBT and SAT positive rates in adult sheep with aerosol immunization were 18.84%higher than oral immunization,while those in lambs were lower.Thus,ae
