摘  要:利用PCR技术对9个斑马源蛔虫线粒体cox1基因部分序列(pcox1)和nad5基因部分序列(pnad5)进行扩增与测序,并分析其遗传多态性和种系发育关系。结果显示,所获得的线粒体pcox1和pnad5序列长度分别为420 bp和528 bp,序列同源性分别为99.3%~100.0%(8个可变位点)和98.1%~99.5%(14个可变位点),检测到单倍型分别为6个和8个,平均单倍型多样性分别为1.834和4.071。系统进化分析结果显示,9个斑马源蛔虫分离株与已知马副蛔虫和Parascaris univalens分离株聚为同一分支,与其他蛔虫所属分支相隔较远,提示马副蛔虫可能与P.univalens为同种异名,但需要进一步研究证实。结果表明,本次试验获得的斑马源蛔虫为马副蛔虫或P.univalens,其线粒体pcox1和pnad5基因均存在一定变异,但pcox1基因变异程度低于pnad5,本试验为斑马蛔虫的分子分类与群体遗传学等研究提供了科学依据。In this study, PCR method was employed to amplify the mitochondrial partial cox1 gene(pcox1) and nad5 gene(pnad5) sequences of 9 Ascaris isolates from zebra in Hunan province, and their genetic polymorphisms and phylogenetic relationships with other Ascaris spp. were analyzed. The results showed that the length of pcox1 and pnad5 sequences were 420 bp and 528 bp, respectively, and the homology of pcox1 and pnad5 sequences were 99.3%-100.0%(8 variable sites) and 98.1%-99.5%(14 variable sites), respectively. In addition, six and eight haplotypes were detected, and the average haplotype diversity for pcox1 and pnad5 sequences were 1.834 and 4.071, respectively. Further phylogenetic analysis showed that 9 Ascaris isolates from zebra gathered in the same branch with Parascaris equorum and Parascaris univalens, and appeared to be far away from the branches of other Ascaris spp., suggesting that P. equorum and P. univalens may belong to the same species, and their taxonomical status requires further validation. These results support the tentative classification of zebra derived Ascaris isolates as P. equorum or P. univalens, which demonstrate low genetic variations in mitochondrial pcox1 and pnad5 sequences, with the variation in pcox1 being lower than that in pnad5. This study highlights the value of targeting mitochondrial genes for molecular phylogeny and population genetics of Ascaris from zebra.
