炒白芍配方颗粒制备工艺优化

摘　　要：目的优化炒白芍配方颗粒的提取、浓缩、干燥和制粒工艺,得到稳定、质量合格的配方颗粒剂。方法以芍药苷含量和得膏率为考察指标,采用正交试验优选炒白芍的水提取工艺;以浸膏性状、芍药苷含量优选浓缩、干燥工艺,以颗粒的成形性优选糊精用量及干法制粒工艺参数,获得最佳制备工艺。结果优化后的炒白芍配方颗粒制备工艺为炒白芍饮片加水煎煮2次,每次加8倍量水,每次煎煮1 h;60℃减压浓缩至相对密度为1.16~1.20;75℃下减压干燥制成浸膏粉,再加适量糊精,混匀后干法制粒(挤压压力为1.5 kgf、送料频率为30 Hz、压片频率为30 Hz),制得炒白芍配方颗粒成品。结论优选的制备工艺简单、稳定、可行,适合大规模生产。Objective To optimize the process of extraction,concentration,drying and granulation of dispensing granules of fried Paeoniae Radix Alba to obtain a stabilized and qualified preparations.Methods Taking the content of paeoniflorin and yield of solid extracts as evaluation indexs,orthogonal test is used to optimize water extraction process of fried Paeoniae Radix Alba;the process of concentration and drying is screened by property of extracts and content of paeoniflorin,the amount of dextrin and parameters of dry granulation are screened by granules formability.Finally,the optimal preparation process is obtained.Results The optimal preparation process of dispensing granules of fried Paeoniae Radix Alba is as follows as adding 8 folds water,decocting twice with 1 h for each time;vacuum concentration at 60℃and relative concentration density 1.16~1.20;powdered extract prepared by vacuum drying method at 75℃,and mixed with appropriate amount of dextrin to prepare dispensing granules of fried Paeoniae Radix Alba by dry granulation(squeeze pressure is 1.5 kgf,feeding frequency is 30 Hz,tabletting frequency is 30 Hz).Conclusion Optimized preparation technology is simple,stable,feasible and suitable for large-scale production of dispensing granules of fried Paeoniae Radix Alba.
