波棱瓜子总木脂素对慢性酒精性肝损伤大鼠肝组织TGF-β/CTGF表达的影响

摘　　要：目的:探讨波棱瓜子总木脂素对慢性酒精性肝损伤大鼠肝脏TGF-β/CTGF表达的影响及保护作用机制。方法:大鼠分为:模型对照组、水飞蓟素50 mg/kg组、波棱瓜子总木脂素100、200、400 mg/kg组,连续灌胃给药8 w,并同时建立慢性酒精性肝损伤大鼠模型,另设正常对照组。取肝脏,计算大鼠的肝脏系数。通过酶联免疫吸附(Elisa)法检测大鼠血清中转化生长因子-β(TGF-β)、结缔组织生长因子(CTGF)以及肝匀浆超氧化物歧化酶(SOD)的活力,观察大鼠肝组织病理切片胶原纤维染色(Masson)结果,免疫组织化学染色观察TGF-β、CTGF蛋白表达情况。结果:与正常对照组相比,肝脏组织经过Masson三色胶原染色后发现,模型对照组有胶原纤维的生成,肝脏系数显著升高,血清中TGF-β、CTGF含量显著升高,肝组织中SOD活力显著升高,肝组织中TGF-β、CTGF蛋白表达显著上调(P<0.01)。与模型对照组相比较,波棱瓜子总木脂素400 mg/kg组大鼠的肝脏系数显著降低(P<0.01),血清中TGF-β、CTGF含量显著降低,肝匀浆SOD活力显著增高(P<0.01),肝脏损伤程度减轻,胶原纤维的生成减少,肝组织中TGF-β、CTGF蛋白表达明显下调(P<0.05或P<0.01)。结论:波棱瓜子总木脂素可以通过抑制CTGF的生成,对酒精性肝损伤大鼠的肝组织具有一定的保护作用。Objective:The aim of this paper was to explore the effect of total lignans in Herpetospermum caudigerum Wall on the expression of TGF-β/CTGF in the livers of rats with chronic alcoholic liver injury, and to discuss its protective mechanism. Methods: A rat model chronic alcoholic liver injury was established, 400 mg/kg, 200 mg/kg, and 100 mg/kg total lignans in Herpetospermum caudigerum Wall were orally administered for eight weeks, the liver tissues were taken to calculate the liver coefficient. The contents of transforming growth factor-β(TGF-β),connective tissue growth factor(CTGF)and superoxide dismutase(SOD)in the liver homogenate were detected by Elisa method, the pathological changes of liver tissues were observed by Masson collagen, and the expressions of TGF-β and CTGF were observed by immunohistochemical staining. Results: Compared with the control group, after the liver tissues were stained with Masson’s three-color collagen, collagen fibers appeared in the model group, the liver coefficient was significantly increased, the expressions of TGF-β and CTGF were significantly up regulated(P<0.01). Compared with the model group, the liver coefficient in 400 mg/kg total lignans group was significantly reduced(P<0.01), the serum contents of TGF-β and CTGF were significantly reduced, the SOD activity in liver homogenate was significantly increased(P<0.01). The liver damage was ameliorated, and the production of collagen fibers was reduced, the expressions of TGF-β and CTGF were significantly down regulated(P<0.05 or P<0.01). Conclusion: The total lignans of Herpetospermum caudigerum Wall can inhibit the production of CTGF and have a certain protective effect on the liver tissues of alcoholic liver injury rats.
