摘 要:以紫花槭(Acer pseudosieboldianum)当年生带腋芽的嫩茎为外植体,研究不同的消毒方法及不同浓度的植物生长调节剂对茎段腋芽诱导、丛生芽增殖和生根的影响,初步建立了紫花槭的快速繁殖体系。结果表明:最适合紫花槭茎段的消毒方法为75%乙醇消毒40 s后再用0.1%HgCl_(2)消毒8 min;腋芽诱导的最佳启动培养基为MS+0.4 mg·L^(-1)吲哚丁酸(IBA),诱导率为93.33%;最佳丛生芽增殖培养基为MS+0.4mg·L^(-1)IBA+0.8 mg·L^(-1)氯吡苯脲(CPPU)+0.6 mg·L^(-1)6-苄氨基嘌呤(6-BA),增殖系数为3.87;最佳生根培养基为1/2MS+0.1 mg·L^(-1)IBA,生根率为48.33%。The young stem segments of Acer pseudosieboldianum were taken as explants,and different sterilization methods were used to compare the sterilization effect of external implants.The MS medium was used as basal medium and the rapid propagation system was established to study the factors influencing axillary buds induction,cluster bud proliferation and rooting culture.The results showed that the most suitable disinfection method for the stems of A.pseudosieboldianum were 75%ethyl alcohol solution for 40 seconds and 0.1%HgCl_(2)solution for 8 minutes.The optimal medium for axillary bud induction was MS+0.4 mg·L^(-1)IBA,with a maximal induction rate of 93.33%.The optimal proliferation medium was MS+0.4 mg·L^(-1)IBA+0.8 mg·L^(-1)CPPU+0.6 mg·L^(-1)6-BA,and the proliferation coefficient was 3.87.The optimal medium for rooting was 1/2 MS+0.1 mg·L^(-1)IBA,and the rooting rate reached 48.33%.
