摘  要:利用流式细胞仪检测大麦(Hordeum vulgare)小孢子离体培养获得的愈伤组织和再生植株细胞核DNA倍性水平及其分布规律,结果表明:大麦‘花30’和‘甘啤1405’小孢子愈伤组织细胞在诱导培养第6天时已发生自发加倍,二倍体细胞数目随着诱导培养时间的延长表现出逐步增加趋势。后期分化时,单倍体、二倍体和四倍体再生植株所占比率分别为7.8%、85.5%、6.7%。两份材料的愈伤产量无明显差异,绿苗分化频率差异显著。结论:大麦小孢子愈伤组织细胞的自发加倍发生在多细胞结构形成(第6天)或更早时期;愈伤分化形成的不同倍性再生植株所占比率相对稳定,不受基因型影响。Flow cytometry was used to detect the ploidy level and distribution of nucleus DNA in calli and regenerated plantlets of barley(Hordeum vulgare)microspore culture in vitro.The results show that microspore-derived callus cells of barley‘Hua 30’and‘Ganpi 1405’doubled spontaneously at 6th day of induction,and the amount of doubled cells showed a gradual increasing trend with the extension of induction.The average percentages of haploid,diploid and tetraploid plantlets were 7.8%,85.5%and 6.7%,respectively,at the later stage of differentiation.There was no significant difference in callus yield between the two materials,but there was a significant difference in green plantlet differentiation frequency.In conclusion,the spontaneous doubling of barley microspore-derived callus cells occurs at the stage of multi-cellular structure formation(6th day)or earlier.The percentage of different ploidy-regenerated plantlets from calli differentiation was relatively stable,which was not affected by genotype.
