摘 要:[目的]了解香蕉CASP家族成员的生物学功能。[方法]以拟南芥CASP基因作为参考序列,基于香蕉基因组数据,通过Blast鉴定香蕉A、B基因组中的CASP家族成员,并对CASP家族成员进行序列特征及低温下的表达分析。[结果]共鉴定得到61个香蕉CASP家族成员,其中A基因组44个(MaCASP)、B基因组17个(MbCASP);CASP基因编码的蛋白存在DUF588和MARVEL保守结构域;根据与拟南芥的亲缘关系将香蕉A、B基因组中的CASP家族成员分为5类;CASP家族成员启动子顺式作用元件中包含光响应元件、激素响应元件、压力响应元件以及多种生长发育响应元件;在低温胁迫下,6个成员(MaCASP21、MaCASP19、MaCASP32、MaCASP43、MaCASP11、MaCASP16)在4℃时呈现特异性表达。[结论]香蕉CASP家族具有功能的多样性,且CASP家族成员在A、B基因组中存在一定的差异,还可能参与低温响应过程。[Purpose]To understand the biological functions of banana CASP family.[Method]In this study,the CASP gene family members from banana A and B genomes were identified by BLAST using Arabidopsis CASP gene as the reference sequence.The CASP gene family members were further analyzed for sequence characterization and expression at low temperatures.[Result]A total of 61 banana CASP family members were identified,including 44 in the A genome(MaCASP)and 17 in the B genome(MbCASP).All the proteins encoded by the CASP genes consisted of 2 conserved structural domains,DUF588 and MARVEL.The CASP gene family members in banana A and B genomes were classified into 5 groups according to their affinities with those of Arabidopsis.The cis-acting elements in the promoters of CASP gene family members included light-responsive elements,hormone-responsive elements,stress-responsive elements and multiple elements in response to growth and development.Under low temperature stress,6 gene members(MaCASP21,MaCASP19,MaCASP32,MaCASP43,MaCASP11,and MaCASP16)were specifically expressed at 4℃.[Conclusion]The banana CASP gene family were functionally diverse,and they were somewhat different in the A and B genomes,which might be involved in the gene regulation under low temperature stress.
