韭菜迟眼蕈蚊雌雄成虫差异基因表达的转录组分析

摘  要:为明确韭菜迟眼蕈蚊(Bradysia cellarum)雌雄成虫之间存在的差异基因及其表达与功能富集,分别对韭菜迟眼蕈蚊雌雄成虫进行全长转录组测序,利用R语言DESeq包比较它们之间的基因表达差异,然后进行KEGG富集分析。结果表明,韭菜迟眼蕈蚊雄性的基因在数量上多于雌性,其中雄性特有的基因6833个,雌性特有的基因3898个,两性共有的基因17763个;雌雄成虫差异表达基因共18675个,其中在雌性中表达上调基因9794个,表达下调基因8881个;差异表达基因明显富集在有机酸代谢、肽代谢过程等功能类别上,参与核糖体生物合成、RNA转运等KEGG通路。研究结果为揭示韭菜迟眼蕈蚊的基因功能、明确基因间的相互关系以及性别决定与性别分化的分子机理提供了基础数据。To clarify the differential gene expression and functional enrichment between male and female adults of Bradysia cellarum,the full-length transcriptome sequencing was performed on male and female adults of Bradysia cellarum,and the differences of gene expression between them were compared by using DESeq package of R language,and then KEGG enrichment analysis was performed.Results show that the number of male genes of Bradysia cellarum is more than that of female genes,including 6833 male specific genes,3898 female specific genes and 17763 genes shared by both sexes;There are 18675 differentially expressed genes in male and female adults,including 9794 up-regulated genes and 8881 down-regulated genes in females;The differentially expressed genes are significantly enriched in organic acid metabolism,peptide metabolism,etc.,and are involved in the KEGG pathways,such as ribosome biogenesis and RNA transport,etc.The results provides the basic data to reveal the gene function of Bradysia cellarum,and clarify the interrelationship between genes and the molecular mechanism of sex determination and sex differentiation.
