摘 要:该研究通过对比蛋鸡不同生长发育阶段卵巢组织,筛选鉴定与蛋鸡繁殖性能相关的长链非编码RNA(long non-coding RNA,lncRNA)。通过提取20周龄(n=3)、30周龄(n=3)京红蛋鸡卵巢组织总RNA构建测序文库,利用RNA-seq技术及生物信息学分析方法鉴定差异表达lncRNA并进行qRT-PCR验证及功能富集分析。在不同周龄蛋鸡卵巢组织中共筛选鉴定到1283个lncRNA表达,其中10个存在差异表达,qRT-PCR验证了测序结果。差异表达lncRNA显著富集到190条GO条目及5条KEGG通路中。研究结果表明lncRNA参与蛋鸡繁殖性能调控相关的生物学过程、细胞组分、分子功能以及多种信号通路。In this research,the lncRNA in different growth and development stages were identified and their relationship with production traits of laying hens was explored.The total RNA in ovarian tissues were extracted from JingHong laying hens in different development stages(20 week n=3 and 30week n=3).Then RNA-seq were carried out to identify differentially expressed lncRNAs.qRT-PCR was used to verify RNA-seq result.GO and KEGG were carried out to predict the function of lncRNA.As a result,a total of 1283 lncRNA in different ovarian tissues were identified,among them,10 were differentially expressed.qRT-PCR verified the sequencing results.Functional enrichment of differential expression of lncRNA showed significant enrichment in 190 GO entries and 5 KEGG pathways.This study showed that lncRNA participates in the breeding performance regulation of laying hens such as regulating biological processes,cell components,molecular function,and multiple signaling pathways.
