摘 要:目的:探讨三七总皂苷通过B淋巴细胞瘤-2(Bcl-2)/Bcl-2相关x蛋白(Bax)/含半胱氨酸的天冬氨酸蛋白水解酶-3(Caspase-3)信号通路对自身免疫性心肌炎(EAM)模型大鼠心肌凋亡的影响。方法:55只Lewis大鼠被分成对照组(n=11)和造模组(n=44)。造模组采用猪心肌肌球蛋白乳浊液后足垫皮下注射建立EAM模型,对照组大鼠给予等体积完全弗氏佐剂后足垫皮下注射。成功建立EAM模型的大鼠被分成模型组、三七总皂苷低、高剂量组和甲氨蝶呤组各11只。模型组、对照组通过灌胃给予蒸馏水10 ml/kg,每日1次;三七总皂苷低、高剂量组分别灌胃给予10、20 mg/ml三七总皂苷溶液10 ml/kg,每日1次;甲氨蝶呤组灌胃给予0.1 mg/ml甲氨蝶呤混悬液10 ml/kg,每周1次。各组大鼠均给药4周。结果:与模型组比较,三七总皂苷低、高剂量组和甲氨蝶呤组大鼠左室射血分数(LVEF)和左室短轴缩短率(LVFS)较高,而左室舒张末期内径(LVIDd)、左室收缩末期内径(LVIDs)水平较低,心肌组织病理评分较低,心肌组织细胞凋亡率较低,心肌组织Bcl-2表达水平较高,Bax、Caspase-3表达水平较低(均P<0.05),且三七总皂苷干预组呈现一定量效关系。结论:三七总皂苷能剂量依赖性的改善EAM大鼠心功能,减轻心肌炎症病理变化,抑制细胞凋亡,其作用可能与调节Bcl-2/Bax/Caspase-3信号通路中凋亡相关蛋白表达有关。Objective:To analysis the effect of panax notoginseng saponins on myocardial apoptosis in rats with experimental autoimmune myocarditis(EAM)based on B-lymphoma-2(Bcl-2)/Bcl-2-related x protein(Bax)/cysteine containing aspartate protein hydrolase-3(Caspase-3)signaling pathway.Methods:Fifty-five Lewis rats were divided into control group(n=11)and model building group(n=44).The model building group was established EAM model by subcutaneous injection of porcine myosin emulsion into the foot pad,rats in the control group received subcutaneous injection of equal volume complete Freund’s adjuvant into the foot pad.After the model was established,the rats in model building group were randomly divided into model group,panax notoginseng saponins low-dose,high-dose group and methotrexate group,each with 11 rats.The model group and control group were given distilled water 10 ml/kg respectively by gavage,once a day;panax notoginseng saponins low-dose,high-dose group were given 10,20 mg/ml panax notoginseng saponins solution 10 ml/kg respectively by gavage,once a day;methotrexate group was given methotrexate suspension 10 ml/kg by gavage,once a week.All rats in each group were treated for 4 weeks.Results:Compared with model group,left ventricular ejection fraction(LVEF)and left ventricular short axis shortening rate(LVFS)were higher,the levels of left ventricular end diastolic diameter(LVIDd)and left ventricular end systolic diameter(LVIDs)were lower,the pathological scores of myocardial tissue were lower,the apoptosis rates of myocardial tissue were lower,the expression levels of Bcl-2 in myocardium were higher,and the expression levels of Bax and Caspase-3 were lower in panax notoginseng saponins low-dose,high-dose group and methotrexate group(all P<0.05),and panax notoginseng saponins intervention group showed a certain dose effect relationship.Conclusion:Panax notoginseng saponins can can improve the cardiac function of EAM rats in a dose-dependent manner,alleviate the pathological changes of myocarditis and inhibit apop
