摘  要:目的探究泻肺逐饮汤对肺癌胸水大鼠胸膜组织细胞角蛋白片段21-1(CYFRA211)、转化生长因子(TGF-β1)、α1酸性糖蛋白(α1-AGP)的影响。方法选取60只SD大鼠,采用随机数字表法分为:健康对照组、模型组、低剂量泻肺逐饮汤组和高剂量泻肺逐饮汤组,每组15只。除健康对照组外各组大鼠种植艾氏腹水瘤细胞制成肺癌胸水模型,低剂量和高剂量泻肺逐饮汤组采用泻肺逐饮汤灌胃处理,作用剂量分别为16.1 g/(kg·d)和32.2 g/(kg·d),1次/d,持续7 d。每组选取6只大鼠,比较各组生存期;剩余大鼠完成治疗后取胸水,比较各组大鼠胸水量;采用HE染色观察胸膜组织和胸水沉渣细胞形态;采用蛋白质印迹法检测各组大鼠胸膜组织中CYFRA211、TGF-β1、α1-AGP的表达情况。结果相比健康对照组,模型组大鼠平均生存时间、中位生存期均显著降低,胸水量、CYFRA211、TGF-β1和α1-AGP蛋白的相对表达水平均显著升高(P<0.05);相比模型组,低剂量泻肺逐饮汤组大鼠平均生存时间、中位生存期均显著升高,胸水量、CYFRA211、TGF-β1和α1-AGP蛋白的相对表达水平均显著降低(P<0.05);相比低剂量泻肺逐饮汤组,高剂量泻肺逐饮汤组大鼠平均生存时间、中位生存期均显著升高,胸水量、CYFRA211、TGF-β1和α1-AGP蛋白的相对表达水平均显著降低(P<0.05)。膜组织和胸水沉渣染色观察发现,模型组大鼠胸膜明显增厚,炎性浸润严重,且有大量癌细胞聚集形成不规则形状;使用泻肺逐饮汤处理后大鼠胸膜增厚明和细胞炎性浸润显著改善,且聚集形成不规则图装癌细胞显著减少,高剂量泻肺逐饮汤组效果优于低剂量泻肺逐饮汤组。结论泻肺逐饮汤可以显著降低大鼠胸膜组织CYFRA211、TGF-β1、α1-AGP的表达。Objective To explore the effects of Xiefei Zhuyin Decoction(泻肺逐饮汤) on cytokeratin fragment 21-1(CYFRA211), transforming growth factor(TGF-β1) and α1 acid glycoprotein(α1-AGP) in pleural tissues of rats with lung cancer pleural effusion. Methods Sixty SD rats were enrolled and divided into healthy control group, model group, low-dose and high-dose Xiefei Zhuyin Decoction groups by random number table method, 15 cases in each group. Except for healthy control group, Ehrlich ascites tumor cells(EATCs) were planted to prepare lung cancer pleural effusion models in the other groups. The low-dose and high-dose Xiefei Zhuyin Decoction groups given Xiefei Zhuyin Decoction intragastrically with the dose of 16.1 g/(kg·d) and 32.2 g/(kg·d)(once/d for 7 d), respectively. Six rats in each group were selected to compare their survival. The pleural effusion was collected from the remaining rats after treatment. The pleural effusion volume was compared among all groups. The cells morphology in pleural tissues and pleural fluid sediment was observed by HE staining. The expressions of CYFRA211, TGF-β1 and α1-AGP in pleural tissues of each group were detected by Western blot. Results Compared with those of the healthy control group, the average survival time and median survival time were significantly shortened, while pleural effusion volume and relative expression levels of CYFRA211, TGF-β1 and α1-AGP proteins were significantly increased in the model group(P<0.05). Compared with those of the model group, the average survival time and median survival time were significantly prolonged, while pleural effusion volume and relative expression levels of CYFRA211, TGF-β1 and α1-AGP proteins were significantly decreased in the low-dose Xiefei Zhuyin Decoction group(P<0.05).
