自拟固胎煎对复发性流产小鼠蜕膜组织Foxp3、RORγt表达的影响

摘　　要：目的探讨自拟固胎煎对复发性流产小鼠蜕膜组织中叉头/翼状螺旋转录因子3(forkhead/pterygoid spiral transcription factor 3,Foxp3)、维甲酸相关核孤儿受体(retinoic acid related nuclear orphan receptor,RORγt)表达的影响。方法将CBA/J雌鼠与BALB/c雄鼠2∶1合笼,建立正常妊娠模型(CBA/J×BALB/c组)作为对照组。CBA/J雌鼠与DBA/2雄鼠2∶1合笼,建立复发性流产模型(CBA/J×DBA/2组),并随机分为中药高、中、低剂量组、西药组以及模型组,HE染色观察各组蜕膜细胞形态,免疫组化法及Western Blot法分别检测各组小鼠蜕膜组织中Foxp3和RORγt的表达。结果与模型组相比,自拟固胎煎中、高剂量组小鼠蜕膜组织内细胞数量及血管数量增多,胞质丰富,胞浆水肿减轻,核仁大而明显。且Foxp3表达均显著增加(P<0.05),RORγt的表达均显著降低(P<0.05)。结论自拟固胎煎能增加蜕膜组织Foxp3的表达,减少RORγt的表达,改善小鼠蜕膜组织细胞局部环境,提示自拟固胎煎治疗复发性流产可能与其参与免疫调节,改善蜕膜环境作用机制相关。Objective To explore the effect of self-made Gutai decoction on the expression of Foxp3 and RORγt in the decidua tissue of recurrent spontaneous abortion mice.Methods CBA/J female mice and BALB/C male mice were caged at 2∶1 to establish a normal pregnancy model(CBA/J×BALB/C group)as the control group.CBA/J female and DBA/2 male were caged at 2∶1 to establish the recurrent spontaneous abortion model(CBA/J×DBA/2 group),and randomly divided into the high-dose,medium-dose and low-dose Chinese medicine group,western medicine group and model group.HE staining to observe the morphology of decidual cells in each group,Immunohistochemistry and Western blot were used to detect the expression of Foxp3 and RORγt in decidua of mice in each group.Results Compared with the model group,the number of blood vessels in the medium-dose and high-dose decoction groups increased in the decidual tissue of mice,the cytoplasm was more abundant,the cytoplasm edema was reduced,and the nucleoli were large and obvious;the expression of Foxp3 was significantly increased in the medium dose group and high dose group(P<0.05),and the expression of RORγt was significantly decreased(P<0.05).Conclution Self-made Gutai decoction can increase the expression of Foxp3 and reduce the expression of RORγt,and and improve the local environment of mouse decidua tissue cells.It is suggested that the mechanism of self-made Gutai decoction in treating recurrent spontaneous abortion may be related to its participation in immune regulation and improving decidual environment.
