摘　　要：尖孢炭疽菌是导致丹霞苹果果实软腐的主要病原菌,在细胞水平研究苹果自身对尖孢炭疽菌多聚半乳糖醛酸酶(PG)侵染的抗病能力,是防治苹果果实软腐病的有效途径。试验从丹霞苹果病斑提取、培养尖孢炭疽菌,用琼脂糖径向扩散法和分光光度法测定PG活性;同时从苹果健康组织中提取多聚半乳糖醛酸酶抑制蛋白(PGIP),用琼脂糖径向扩散法及接种试验测定PGIP对PG的抑制效果。结果表明,提取培养的尖孢炭疽菌多聚半乳糖醛酸酶具有良好活性,在培养8~20 h期间酶活性呈线性回归(R2=0.99);琼脂糖径向扩散显示,PGIP抑制PG效果显著,光晕面积缩小65%以上;接种试验表明,PGIP对PG的抑制率在27.9%~54.5%。丹霞苹果果实内部多聚半乳糖醛酸酶抑制蛋白(PGIP)对尖孢炭疽菌多聚半乳糖醛酸酶有明显的抑制作用。Colletotrichum acutatum is the main pathogen causing soft rot of Danxia apple fruit. It is an effective way to control soft rot of apple fruit to study the resistance of apple against polygalacturonase(PG)infection of Colletotrichum acutatum at the cellular level.Colletotrichum acutatum was extracted and cultured from Danxia apple disease spot, and the PG activity was determined by agarose radial diffusion method and spectrophotometry. Polygalacturonase inhibitor protein(PGIP)was extracted from healthy apple tissues. The inhibitory effect of PGIP on PG was measured by agarose radial diffusion method and inoculation test. The results showed that the polygalacturonase extracted and cultured from Colletotrichum acutatum had good activity. The enzyme activity showed a linear regression(R2=0.99)during 8-20 h. Radial diffusion of agarose assay showed that PGIP inhibited PG significantly, and the halo area decreased by more than 65%. Inoculation experiments showed that the inhibition rate of PGIP against PG ranged from 27.9% to 54.5%. In conclusion,the polygalacturonase inhibitory protein(PGIP) in Danxia apple fruits has significant inhibitory effect on polygalacturonase of Colletotrichum acutatum.
