摘　　要：单细胞RNA测序技术是基于新一代测序平台在单细胞水平上进行的转录组分析,具有高通量和高灵敏度等优势。在正常皮肤中,单细胞RNA测序可用于鉴定皮肤细胞的异质性水平,尤其是成纤维细胞和髓源性细胞。该技术还应用于银屑病、特应性皮炎、皮肤肿瘤、系统性硬化症、红斑狼疮等多种皮肤病中,用来揭示细胞异质性、阐明发病机制和确定新的诊断标志物和治疗靶点等。本文对单细胞RNA测序技术及其在皮肤病研究中的应用进行综述,旨在提高皮肤疾病的诊疗水平。Single cell RNA sequencing(scRNA-seq)is a technique to analyze transcriptome sequences at the single cell level based on the new generation sequencing platform.It has the advantages of high throughput and high sensitivity.scRNA-seq can be used to identify cellular heterogeneity in normal skin,particularly fibroblasts and myeloid cells.This technique is also useful to reveal cellular heterogeneity,elucidate the pathogenesis and identify novel diagnostic and therapeutic targets in skin diseases,including psoriasis,atopic dermatitis,skin tumors,systemic sclerosis and lupus erythematosus.This review introduces the scRNA-seq and its application in normal skin and skin diseases,aiming to improve the diagnostic and therapeutic levels of skin diseases.
